窒息对新生小鼠心肌β受体表达的影响

摘  要:目的探讨窒息对新生小鼠心肌β受体表达的影响及β2受体激动剂的干预效果。方法清洁级出生3 d的ICR小鼠108只,雌雄不限,采用随机数字表法分为:正常组(n=36)仅置于广口瓶中,未给予处理;窒息组(n=36)参照文献方法制备新生鼠常压窒息模型;干预组(n=36)处理同窒息组,但在窒息前2 h单次腹腔注射非诺特罗。干预后分别于小鼠出生后3、5、7、14、21、28 d随机从各组取6只,测定心电图,处死,计算心重指数(HW/BW)。留取心脏组织,免疫组化法和蛋白免疫印迹法(Western blot)检测β1、β2受体表达。结果正常小鼠心肌组织β1受体免疫组化染色,可见心肌细胞排列整齐,细胞核清晰,细胞膜完整,无炎性细胞浸润。细胞核膜及胞浆着色,随着小鼠日龄(3~28d)增加,黄染逐渐变深。Western blot结果显示28 d小鼠心肌β1受体相对表达量明显高于3 d的小鼠,差异有统计学意义(P<0.05)。β2受体主要为细胞核膜及胞浆着色,随着日龄增加,着色变浅,Western blot显示28 d小鼠心肌β2受体相对表达量明显低于3 d的小鼠,差异有统计学意义(P<0.05)。与正常组3 d小鼠比较,窒息处理后,小鼠心肌β1、β2受体免疫组化染色变浅,表达均明显下降,且窒息组心肌细胞排列紊乱,有少许炎性细胞渗出。Western blot结果显示,窒息组较正常组心肌β1、β2受体相对表达量降低,差异有统计学意义(P均<0.05)。与正常组28 d小鼠相比,干预组及窒息组小鼠心肌β1、β2受体相对表达量均降低,差异有统计学意义(P均<0.05)。干预组28 d小鼠心肌β1、β2受体相对表达量相比于窒息组有升高趋势。28 d小鼠中,窒息组较正常组HW/BW升高,干预组较窒息组HW/BW降低,差异有统计学意义(P均<0.05)。结论未成熟心肌发育过程中,正常小鼠β1受体表达升高,β2受体表达下降,窒息会导致β1、β2受体表达均下降,而特异性β2受体激动剂非诺特罗预处理,可促进β1、β2受�Objective To investigate the influence of asphyxia on expression of myocardialβ-adrenergicreceptor and interventional effect onβ2-adrenoceptor agonist in neonatal mice.Methods Clean ICR mice(n=108,male or female)born for3d were randomly divided into normal group put into wide-mouth bottlewithout any treatment,asphyxia group prepared to ordinary pressure asphyxia model according to literaturemethod,and intervention group given intraperitoneal injection of fenoterol2h before asphyxia(each n=36).The mice(n=6)were randomly chosen from each group on the3rd d,5th d,7th d,14th d,21st d and28th d fordetecting electrocardiogram(ECG)and calculating heart weight index(HW/BW).The samples of cardiac tissuewere collected for detecting the expressions ofβ1-adrenergic receptor andβ2-adrenergic receptor by usingimmunohistochemistry technique and Western blot.Results The results of immunohistochemistry staining ofβ1-adrenergic receptor in normal mice showed that alignment myocardial cells,distinct nucleus and intact cellmembrane without inflammatory cell infiltration.The staining of cell nuclear envelope and plasma darkened asthe age of mice increased(from3d to28d).The results of Western blot showed that relative expression ofβ1-adrenergic receptor was significantly higher in28-d mice than that in3-d mice(P<0.05).The staining ofβ2-adrenergic receptor showed that staining of cell nuclear envelope and plasma became shallow as the age of miceincreased.The results of Western blot showed that relative expression ofβ2-adrenergic receptor was significantlylower in28d mice than that in3d mice(P<0.05).The staining ofβ1-adrenergic receptor andβ2-adrenergicreceptor became shallow,and expressions of them decreased significantly in asphyxia group compared with normalgroup on the3rd d,and there were malalignment myocardial cells and a few exudate inflammatory cells observed inasphyxia group.The results of Western blot showed that relative expressions ofβ1-adrenergic receptor andβ2-adrenergic receptor decreased in asphyxia group comp
