摘 要:目的分析冠状动脉粥样硬化性心脏病(冠心病)心力衰竭患者检测胸腹水细胞中泛素活化酶(UBE1)、端粒酶的应用价值。方法选取2013年1月~2016年1月于新乡市第一人民医院检验科保存的57例胸腹水标本,其中30例为冠心病心力衰竭,15例为冠心病(非心力衰竭,死因为颅脑外伤或机械性损伤),12例正常对照(无心脏病史、无心脏器质性病变、死因为颅脑外伤或机械性损伤)。免疫组化染色检测各组UBE1和端粒酶表达情况。结果心力衰竭组UBE1免疫组化染色阳性和强阳性比例明显高于冠心病组和正常对照组,差异有统计学意义(P均<0.05)。与冠心病组和正常对照组比较,心力衰竭组端粒酶免疫组化染色阳性和强阳性比例明显升高,差异有统计学意义(P均<0.05)。心力衰竭组UBE1和端粒酶IOD值分别为1201.14(943.19~1672.31)和12 643.81(11 231.07~13 519.31),明显高于冠心病组和正常对照组,差异有统计学意义(P均<0.05)。结论冠心病心力衰竭患者胸腹水细胞中泛素活化酶、端粒酶表达明显增强,二者在冠心病心力衰竭诊断中有潜在的应用价值。Objective To analyze the application values of detection of ubiquitin activating enzyme E1(UBE1)and telomerase in pleural-peritoneal fluid cells in patients with heart failure of coronary heart disease(CHD).Methods The samples of pleural-peritoneal fluid from57patients saved in the Department of ClinicalLaboratories of the First People’s Hospital of Xinxiang City were chosen from Jan.2013to Jan.2016.Allsamples were divided into heart failure group(n=30)collected from the patients with grades fromⅢtoⅣcoronarystenosis(excluding other death causes),CHD group(n=15)from the patients with grades from I toⅣcoronarystenosis(without heart failure,and death causes being craniocerebral trauma or mechanical injury)and normalcontrol group(n=12)from the patients without heart disease history and cardiac organic lesions,and death causesbeing craniocerebral trauma or mechanical injury.The expressions of UBE1and telomerase were detected byusing immunohistochemistry staining method in all groups.Results The percentages of UBE1with positive andstrongly positive immunohistochemistry staining were significantly higher in heart failure group than those in CHDgroup and normal control group(all P<0.05).The percentages of telomerase with positive and strongly positiveimmunohistochemistry staining increased significantly in heart failure group compared with CHD group and normalcontrol group(all P<0.05).The value of integral optical density(IOD)of UBE1was1201.14(943.19~1672.31)andthat of telomerase was12643.81(11231.07~13519.31)in heart failure group,which were significantly higher thanthose in CHD group and normal control group(all P<0.05).Conclusion The expressions of UBE1and telomeraseincrease significantly in pleural-peritoneal fluid cells in patients with CHD heart failure,and they have potentialapplication values in diagnosis of CHD heart failure.
