摘  要:从河南、甘肃和新疆等3个地区采集表现疑似病毒病症状的西瓜和甜瓜叶片样品共132份,采用双抗体夹心酶联免疫吸附(DAS-ELISA)法和反转录PCR(RT-PCR)对其进行病毒检测,以期了解这3个地区的西瓜甜瓜病毒种类及分布,旨在为当地西瓜和甜瓜病毒病的防控提供参考。结果表明:瓜类蚜传黄化病毒(Cucurbit aphid-borne yellows virus,CABYV)的检出率最高,为57.1%,其次为小西葫芦黄花叶病毒(Zucchini yellow mosaic virus,ZYMV)和西瓜花叶病毒(Watermelon mosaic virus,WMV),检出率分别为45.6%、38.9%,黄瓜花叶病毒(Cucumber mosaic virus,CMV)和甜瓜蚜传黄化病毒(Melon aphid-borne yellows virus,MABYV)的检出率较低,分别为20.0%、17.5%,而番木瓜环斑病毒西瓜株系(Papaya ring spot virus-watermelon strain,PRSV-W)和南瓜花叶病毒(Squash mosaic virus,Sq MV)未检出,河南和新疆发现MABYV尚属首次。本试验发现在河南通许危害西瓜甜瓜的病毒有ZYMV、WMV、CABYV、MABYV,在甘肃瓜州为ZYMV、CMV、CABYV,在新疆鄯善为ZYMV、WMV、CMV、CABYV、MABYV。其中21.2%的样品受到2种病毒的复合侵染。另外,3个地区的病毒优势种各不相同,鄯善县的优势种为CABYV,通许县的优势种是WMV,瓜州县的优势种是CMV。132samples with suspected virus-like symptoms were collected from Henan,Gansu Provincesand Xinjiang Autonomous Region.Virus detection was carried out by Double Antibody Sandwich EnzymeLinked Immunosorbent Assay(DAS-ELISA)and Reverse transcript polymerase chain reaction(RT-PCR),hoping to understand the virus species infecting watermelon and melon grown in these3areas and their distribution.The results showed that the detection rates of Cucurbit aphid-borne yellows virus(CABYV),Zucchini yellowmosaic virus(ZYMV),Watermelon mosaic virus(WMV),Cucumber mosaic virus(CMV),Melon aphidborneyellows virus(MABYV)among the132detected samples were57.1%,45.6%,38.9%,20.0%,17.5%,respectively,whereas Papaya ring spot virus-watermelon strain(PRSV-W)and Squash mosaic virus(SqMV)were not detected.It was the first time to find MABYV in Henan Province and Xinjiang AutonomousRegion.This study also found that ZYMV,WMV,CABYV,MABYV occurred in Henan Province;ZYMV,CMV,CABYV in Gansu Province;ZYMV,WMV,CMV,CABYV,MABYV in Xinjiang Autonomous Region.21.2%of these samples were mix-infected by2viruses.Besides,the virus dominant species were different fromeach other in these3areas,implying the distributions were associated with environmental condition and cultivarsgrown.
