摘  要:目的 HSYA与芍药苷联合用药对急性局灶性脑缺血再灌注损伤大鼠脑组织与血小板活化的影响。方法 SPF级雄性SD大鼠,随机分为HSYA组(5.0 mg·kg^(-1))、芍药苷组(5.0 mg·kg^(-1))HSYA与芍药苷联合用药组(合用组)(各5.0 mg·kg^(-1))、银杏内酯组(5.0 mg·kg^(-1))、模型组和假手术组。大脑中动脉线栓法复制脑缺血再灌注损伤模型。脑缺血1 h再灌注6 h后尾静脉注射给药7 d,末次给药2 h后收集相应标本保存;苏木精-伊红(HE)染色法观察海马区神经细胞形态变化;流式细胞仪(FCM)法检测全血中P-选择素(CD62P)含量。结果 HE染色结果提示,与模型组比较,HSYA、芍药苷及合用组均能明显改善急性脑缺血再灌注大鼠海马区神经细胞的损伤程度,以合用组尤为明显。FCM结果提示,与假手术组比较,模型组大鼠全血中CD62P的活化程度显著增加(p<0.01);与模型组相比,合用组与银杏内酯组均能显著抑制全血中CD62P的活化程度(p<0.05);与合用组相比,芍药苷组全血中CD62P的活化程度显著增加(p<0.01),但HSYA组与合用组之间抑制作用无显著性差异。结论 HSYA与芍药苷联合用药对脑缺血再灌注损伤模型大鼠的的海马区神经细胞保护机制可能与抑制CD62P表达有关。Objective To investigate the effects of HSYA combined with paeoniflorin on brain tissue and the expression of platelet activation in rat models with cerebral ischemia-reperfusion injury.Methods Male SPF of SD rats were randomly divided into HSYA group(5mg*kg-1),paeoniflorin group(5mg*kg-1),combination group HSYA and paeoniflorin(combination group)(5mg*kg-1)and ginkgolide group(5mg*kg-1),model group and the sham operation group.The rat model of middle cerebral artery occlusion(MCAO)was induced by modified suture method.After1h of cerebral ischemia followed by6h reperfusion,rats tail intravenous injection for7days,to collect and preserve the corresponding index after the last administration of2h;hematoxylin eosin(HE)staining was used to observe the morphological changes of nerve cells in the hippocampus;flow cytometry(FCM)assay was used to observe the P-selectin(CD62P)content in blood.Results The results of HE staining showed that compared with the model group,HSYA,paeoniflorin and the combination group could obviously improve the damage degree of the neurons in the hippocampus of rats with acute cerebral ischemia and reperfusion.FCM results showed that compared with sham operation group,the activation degree of CD62P was significantly increased in model group(p<0.01);compared with the model group,combination group and the degree of activation of ginkgolide groups could significantly inhibit the whole blood CD62P^<0.05);compared with the combined group,the activation degree of CD62P significantly increased in paeoniflorin group(p<0.01),but between the HSYA group and the combination group had no significant difference inhibition.Conclusion The protective mechanism of HSYA combined with paeoniflorin on hippocampal neurons may be related to inhibition of CD62P expression on cerebral ischemia-reperfusion injury in rats.
