摘 要:目的探讨神经干细胞移植对脑外伤大鼠神经功能及脑组织Caspase-3和Caspase-9表达的影响。方法108只SD成年大鼠随机分为假手术(Sham)组、脑外伤(TBI)组和神经干细胞移植(NSC)组,每组36只。TBI和NSC组采用改良的Feeney氏法制作大鼠脑外伤动物模型,NSC组大鼠在脑外伤模型制作成功后1周在皮质处局部注射移植神经干细胞,TBI和NSC组大鼠分别在细胞移植后的第7天、14天和21天进行神经功能缺损评分(NSS);sham组仅切开头皮暴露颅骨,不实施打击。免疫组织化学方法和Western blot方法检测各组大鼠脑组织Caspase-3和Caspase-9蛋白的表达。结果各时点NSC组大鼠较TBI组NSS评分均降低(P<0.01)。免疫组织化学方法和Western blot均显示,与Sham组相比,TBI组大鼠脑组织的Caspase-3和Caspase-9阳性表达OD值均升高(P<0.05);而与相同时间点的TBI组大鼠相比,NSC组大鼠脑组织Caspase-3和Caspase-9的阳性表达OD值均降低,但仍高于Sham组(P<0.05)。结论神经干细胞移植后可能通过下调Caspase-3和Caspase-9的表达促进脑外伤大鼠神经功能的恢复。Objective To study the effect of neural stem cell transplantation on neurological behavior and expressionsof Caspase-3and Caspase-9in rats with traumatic brain injury(TBI).Methods A total of108SD rats were randomlydivided into the sham group(n=36),traumatic brain injury group(TBI,n=36)and neural stem cell transplantation group(NSC,n=36).TBI model of rats was established by using improved Feeney method.Rats of NSC group were injected neuralstem cells in cortex after traumatic brain injury was established for1week,and then nurological severity scores(NSS)of ratswere evaluated at7days,14days and21days after cell transplantation in TBI and NSC groups,respectively.Rats of shamgroup were only cut the scalp and exposed the skull without striking.The expressions of Caspase-3and Caspase-9in braintissues were detected by immunohistochemistry and Western blot method.Results Compared with NSC group at the sametime points,NSS was decreased significantly in TBI group(P<0.01).Compared with the sham group,the optical density(OD)values of Caspase-3and Caspase-9positive expressions were increased significantly in TBI group(P<0.05).Compared withTBI group at the same time points,the OD values of Caspase-3and Caspase-9positive expressions were decreasedsignificantly in NSC group(P<0.05).Conclusion The neural stem cell transplantation can promote the recovery of braininjury by down-regulating the expressions of Caspase-3and Caspase-9.
