摘 要:目的:探讨肺积方扶正组份对中晚期肺癌患者自体细胞因子诱导的杀伤细胞增殖及杀瘤活性的影响。方法:选取中晚期肺癌患者30例,抽取外周血并分离单个核细胞,在常规加入CIK细胞时,实验组加入肺积方扶正组份含药血清,对照组加生理盐水小鼠血清。检测并比较两组CIK细胞免疫表型、CIK细胞杀伤活性。结果:(1)在细胞培养的第14天,实验组和对照组细胞中CD3^+、CD56^+细胞百分比比较有差异,P<0.05。(2)效靶比为5∶1时,实验组和对照组CIK细胞杀伤活性分别是(13.42±3.74)%和(12.78±3.75)%,P>0.05;效靶比为10∶1时,实验组和对照组CIK细胞杀伤活性分别是(24.42±4.67)%和(21.89±5.32)%,P<0.01;效靶比为20∶1时,实验组和对照组CIK细胞杀伤活性分别是(54.65±8.15)%和(50.06±9.12)%,P<0.05。结论:肺积方拆方之扶正组份天冬、薏苡仁、北沙参有助于提高CIK细胞增殖及杀瘤活性,可能在提高免疫力中起重要作用。Objective:To investigate the effects of Fuzheng component in the Feiji prescription on proliferation and cytotoxicity of autologous CIK cells in patients with advanced lung cancer.Methods:30patients with advanced lung cancer were selected.Peripheral blood was collected and the PBMC were isolated.When the CIK cells were inoculated;the experimental group was treated with serum containing Fuzheng component;and the control group was given saline and mouse serum.The CIK cell immunophenotype and CIK cell killing activity were detected and compared.Results:On the14th day of cell culture,the percentage of CD3+and CD56+cells in the experimental group and the control group was showed a difference(P<0.05).When the effector-target ratio was5∶1,the cytotoxic activity of CIK cells in the experimental group and the control group was(13.42±3.74)%and(12.78±3.75)%respectively,P<0.05.When the effector-target ratio was10∶1,the cytotoxic activity of CIK cells in the experimental group and the control group was(24.42±4.67)%and(21.89±5.32)%respectively,P<0.01.When the effector-target ratio was20∶1,the cytotoxic activity of CIK cells in the experimental group and the control group was(54.65±8.15)%and(50.06±9.12)%respectively,P<0.05.Conclusion:Tiandong,Yiyiren and Beishashen in Fuzheng component in the Feiji prescription can improve the proliferation and killing activity of CIK cells,and may play an important role in improving immunity.
