摘 要:目的建立高同型半胱氨酸血症的动物模型,检测脂肪组织TRB3的表达,探讨TRB3对PI3K/Akt信号通路的影响.方法 20只昆明小鼠分为正常对照组和高同型半胱氨酸血症组,每组10只.饮用含1.5%蛋氨酸3个月建立高同型半胱氨酸血症组模型.2组麻醉处死后取脂肪组织,逆转录PCR检测TRB3、Akt和GLUT4的m RNA表达水平,Western blot检测TRB3、Akt、p-Akt(473)和GLUT4的蛋白质表达水平.结果高同型半胱氨酸血症组的TRB3的m RNA表达较正常对照组增加(P<0.05),Akt和GLUT4的m RNA表达较正常对照组降低,但差异无统计学意义(P>0.05);高同型半胱氨酸血症组的TRB3的蛋白质表达较正常对照组增加(P<0.05),p-Akt(473)蛋白质的表达降低,Akt和GLUT4的蛋白质表达差异无统计学意义(P>0.05).结论同型半胱氨酸可能通过增加脂肪组织TRB3的表达,降低p-Akt(473)和GLUT4蛋白质含量,抑制PI3K/Akt信号通路,影响脂肪组织对葡萄糖的摄取和利用.Objective To build animal model of hyperhomocysteinemia(HHcy),and to investigate theexpression of TRB3in adipose tissue and explore the effect of PI3K/Akt signaling pathway by TRB3.MethodsTwenty mice were divided into the fasting control group(n=10)and the fasting HHcy group(n=10).Normalfood was fed in the fasting control group.Normal food and1.5%(mass fraction)methionine were fed in the fastingHHcy group.After the mice in every group were fed for three months.Adipose tissue in two groups was The mRNAexpressions of TRB3,Akt and glucose transporter4(GLUT4)were detected by reverse transcription PCR.Theprotein expression of TRB3,Akt,p-Akt(473)and GLUT4were measured by Western blot.Results ThemRNA expression of TRB3in the fast HHcy group was increasedas compared with the control group.The mRNAexpression of Akt and GLUT4was decreased,however the result had no significant difference between two groups.Compared with the control group,the protein expression of TRB3was increased and the protein expression of p-Akt(473)was reduced in the fast HHcy group.Whereas the protein expression of Akt and GLUT4had no significantdiffenence between two groups.Conclusion Homocysteine may incease the uptake and utilization of glucose inadipose tissue by inhibiting PI3K/Akt signaling pathway,which may be the results of inducing the expression ofTRB3and downregulating the protein expression of p-Akt(473)and GLUT4.
