摘 要:目的通过香薷传统切制与产地加工炮制一体化工艺的对比,为香薷产地加工炮制一体化技术实施提供可行性依据。方法以软化时间、切制段长、烘干温度作为传统切制工艺的考察因素,以切制段长、烘干温度、烘干时间作为产地加工炮制一体化工艺的考察因素,采用L9(34)正交试验,以香荆芥酚、麝香草酚、挥发油含量为综合评价指标,分别优化传统切制和产地加工炮制一体化工艺,并对2个优选出的工艺进行对比研究。采用Agilent 1200型高效液相色谱仪、ECOSIL-C18色谱柱测定香荆芥酚和麝香草酚含量,以乙腈-0.2%醋酸水溶液为流动相,梯度洗脱,波长274 nm,流速1.0 m L/min,进样量20μL。按照2015年版《中华人民共和国药典》挥发油测定法甲法测定挥发油含量。结果优化的香薷传统切制工艺为:3倍量水淋润软化1 h,切制1.0 cm,50~60℃烘干;优化的香薷产地加工炮制一体化工艺为:趁鲜切制1.0 cm,50~60℃干燥36 h。香薷产地加工炮制一体化最佳工艺条件下测得的香荆芥酚、麝香草酚、挥发油含量均高于传统切制工艺。结论产地加工炮制一体化技术简单方便,能弥补传统切制工艺繁琐、有效成分易流失等不足,切实可行。Objective To provide the feasible basis for the implementation of integration processing technology of origin of Moslae Herba by comparing the traditional cutting processing and integration processing of origin of Moslae Herba.Methods Taking the softening time,the length of cutting section and the drying temperature as the investigation factors of the traditional cutting processing,and the cutting length,drying temperature and drying time as the investigation factors of the integration processing of origin,L9(34)orthogonal test was used.Carvacrol,thymol,volatile oil content were setting as the evaluation indexes,the traditional cutting processing and the integration processing of origin were optimized,and the two selected process were compared.The contents of carvacrol and thymol were deterrmined by Agilent1200high performance liquid chromatography and ECOSIL-C18column.The mobile phase was acetonitrile-0.2%acetic acid,gradient elution,with wavelength of274nm,flow rate of1.0mL/min,sample volume of20μL.The extraction of the volatile oil was in accordance with the volatile oil determination method in Chinese Pharmacopoeia2015edition.Results The optimizedtraditional cutting processing:softening1h with3times amount of water,cutting to1.0cm,drying at50-60℃.The optimized integration processing of origin:cutting to1.0cm in fresh,drying at50-60℃,drying to36h.The contents of carvacrol,thymol,and volatile oil in the optimum integration processing of origin were all higher than the traditional cutting processing.Conclusion The technology of integration processing of origin is simple and convenient,which can make up the defects of the complicated traditional cutting processing and the lessen of effective components,and is feasible.
