抗猪附红细胞体单克隆抗体的制备和鉴定

摘　　要：为了制备特异的抗猪附红细胞体单克隆抗体(monoclonal antibody,MAb),将纯化的猪附红细胞体抗原浓缩后免疫6周龄BALB/c小鼠,通过杂交瘤细胞技术制备MAb。制备的MAb分别通过抗体亚类试剂盒、Western blot进行单抗亚类和特异性检测。结果显示,共获得了2株能稳定分泌MAb的杂交瘤细胞株,命名为3F7和2C4。2株MAb的亚型均为IgG2a,其腹水效价分别可达1:16 000和1:14 000。特异性检测表明,2株MAb均能特异性地与猪附红细胞体结合,而与弓形虫、猪链球菌、猪肺炎支原体和猪繁殖与呼吸综合症病毒均无反应。2株MAb的获得有望为猪附红细胞体病的诊断和防治提供新的材料。In order to generate monoclonal antibodies(MAbs)against Mycoplasma suis,6-week-old BALB/c mice were immunized with purified and concentrated M.suis antigen and antibody secreting hybridomas were obtained by fusing splenocytes and myeloma cells.Two strains of stable hybridoma cell lines secreting monoclonal antibodies were obtained and designated as3F7and2C4.Both strains were IgG2a as determined using the antibody subclass kit.The antibody titers of ascites were up to1:16000and1:14000,respectively.The specificity test showed that both MAbs reacted with M.suis antigen in Western blot but did not recognize antigens of Toxoplasma gondii,Streptococcus suis,Mycoplasma hyopneumoniae and Porcine reproductive and respiratory syndrome virus.The availability of these MAbs against M.suis would provide biological materials for the diagnosis and prevention of eperythrozoonosis.
