清脑片对小鼠镇痛效果的实验研究

摘  要:目的观察清脑片对小鼠尾部压痛及小鼠扭体模型实验的影响。方法 50只小鼠,随机分为5组,每组10只,即清脑片大、中、小剂量组及晕痛定组、生理盐水组。用尾压测痛仪测定小鼠给药前痛阈,将小鼠尾置于压痛仪操作台上,施加连续递增压力,当小鼠出现挣扎退缩或嘶叫反应时,显示器读出的压力值即为痛阈指标。然后分别灌服大、中、小剂量的清脑片混悬液,晕痛定混悬液及同体积生理盐水,连续7 d,于末次给药后1 h测定痛阈值。另取昆明种小鼠50只,随机分为5组,分别灌服大、中、小剂量清脑片混悬液、晕痛定胶囊混悬液及同体积的生理盐水,给药1次/d,连续给药3 d,于末次给药1 h,每只小鼠腹腔注射新配制的0.5%醋酸蒸馏水液0.1 ml/10 g,立即观察小鼠首次出现扭体反应的潜伏期及10 min内的扭体次数。结果清脑片可显著提高小鼠尾根受机械刺激的痛阈,使痛阈提高值显著升高。清脑片可显著延长小鼠首次扭体的潜伏期(P<0.01)、显著减少10 min内扭体次数减少(P<0.01)。结论清脑片对小鼠尾根加压和小鼠扭体模型实验显示有较好的镇痛作用。Objective To observe the effect of Qingnao Tablet on tail tenderness and torque model in mice.Methods Fifty mice were randomly divided into5groups,10mice for each,i.e the larger,medium and smaller dose groups of Qingnao tablets,the positive group(Yuntongding capsule),and the saline group.Tail pressure pain threshold instrument was used for detecting pain threshold before administration of the drug in mice,the mouse tail was placed on the console of the instrument,increasing the pressure continuously,when the mice was struggling back or bray reaction and read-out display pressure value was the pain threshold.Then,the suspension of Qingnao tablet was orally administered in larger,medium and smaller doses of the Qingnao tablets and Yuntongding capsule respectively,and the same volume of normal saline,continuously7d;the pain threshold after the last administration1h was measured.Another50mice were randomly divided into5groups,administered the suspension in larger,medium and smaller doses of the Qingnao tablets and Yuntongding capsule respectively,and the same volume of normal saline,continuously3d.At the last administration of1h,each mouse was injected with0.5%acetic acid distilled water0.1ml/10g,for observing immediately the latency of torque response and the number of torque in10min.Results Qingnao tablets can significantly improve the pain threshold value of mechanical stimulation in tail root,significantly prolonged the latency of first torque in mice(P<0.01),significantly reduce the number of torque times within10min(P<0.01).Conclusion Qingnao tablets has better analgesic efficacy on mouse tail tenderness and torque model in mice.
