青蟹寿组织培养基筛选试验

摘　　要：以青蟹寿母株花剑上的花蕊作为组培材料,以MS培养基为基础,确定最佳增殖和生根培养基成分。试验结果表明:继代培养基配方为1/2MS+6-BA0.6+IBA0.1;生根培养基配方为1/2MS+IBA0.05+NAA0.1,添加琼脂粉6.0 g、蔗糖30.0 g、活性炭粉0.3 g;最佳栽培基质为草炭1份、珍珠岩1/2份、蛭石1/2份、粗砂1/4份。In this experiment,taken the flower buds in stock plant foil of blue crabs as tissue culture material,based on MS medium,the optimal proliferation and rooting medium components were determined.The experimental results showed that:the subculture media component was1/2MS+6-BA0.6+IBA0.1;Rooting media component was1/2MS+IBA0.05+NAA0.1with agar powder added6.0g,sucrose added30.0g and carbon activated added0.3g;The optimal culture medium was1portion of turf,1/2portion of perlite,1/2portion of vermiculite and1/4portion of coarse sand.
