摘 要:目的探讨肾康灵对阿霉素肾病大鼠血小板微颗粒(PMPs)水平的影响。方法在60只健康清洁级雄性Sprague Dawley系大鼠中按简单随机化原则选取15只作为正常组,其余45只作为实验组,采用一次性尾静脉注射阿霉素建立大鼠肾病综合征模型。10 d后随机抽取4只正常组大鼠和6只实验组大鼠处死取血送检。将实验组剩余的36只(造模期间死亡3只)大鼠随机分为病模组、泼尼松组、泼尼松联合肾康灵组,每组12只,分别给予相应药物灌胃;正常组(11只)和病模组以等量蒸馏水灌胃。观察各组给药前后24 h尿蛋白、血清清蛋白及胆固醇、血浆纤维蛋白原的变化,并以流式细胞术检测PMPs水平。结果治疗前实验组大鼠纤维蛋白原、PMPs水平[(3.46±0.25)g/L、(4 410.67±334.31)μL]均明显高于正常组[(1.36±0.20)g/L、(1 230.00±125.16)μL]。第23天泼尼松组大鼠纤维蛋白原、PMPs水平[(3.30±0.29)g/L、(4 350.13±234.17)μL]与病模组[(3.57±0.35)g/L、(4 436.40±341.94)μL]比较,差异均无统计学意义(P>0.05);泼尼松联合肾康灵组大鼠纤维蛋白原、PMPs水平均明显低于泼尼松组和病模组,差异均有统计学意义(P<0.05)。结论肾康灵可明显降低阿霉素肾病大鼠的PMPs水平,改善肾病综合征的血液高凝状态。Objective〓To explore the effect of Shenkangling on platelet microparticles(PMPs)in rats with adriamycin?鄄induced nephropathy(AN).Methods〓In60healthy clean grade male Sprague Dawley rats,15cases were randomly selected as the normal group,the other45cases served as the experimental group.The rat nephropathy syndrome model was established by once tail vein injection of adriamycin.After10d,4cases randomly extracted from the normal group and6cases extracted from the experimental group were killed for collecting blood and sending for detection.The residual36cases in the experimental group(3cases of death during constructing model period)were randomly divided into the AN model group,prednisone group and prednisone+Shenkangling group,12cases in each group.Each group was given corresponding medication gavage.The normal group and AN model group were given the gavage by the equal amount of distilled water.The changes of urinary protein,serum albumin,cholesterol,plasma fibrinogen at24h before and after medication were observed in each group.The level of CD61+PMPs was measured by using the flow cytometry.Results〓The levels of fibrinogen and PMPs before treatment in the experimental group were(3.46±0.25)g/L and(4410.67±334.31)/μL respectively,which were significantly higher than(1.36±0.20)g/L and(1230.00±125.16)μL in the normal group.The levels of fibrinogen and PMPs after3weeks of treatment in the prednisone group were(3.30±0.29)g/L and(4350.13+234.17)/μL,which in the AN model group were(3.57±0.35)g/L and(4436.40±341.94)μL,respectively,the differences were not statistically significant(P>0.05).The levels of fibrinogen and PMPs in the prednisone+Shenkangling group were significantly lower than those in the prednisone group and AN model group,the difference was statistically significant(P<0.05).Conclusion〓Shenkangling can significantly decrease the level of PMPs in rats with AN and improves the hypercoagulability state.〓
