摘  要:目的:研究瘢痕疙瘩中缺氧诱导因子-1α(HIF-1α)的表达及其与血管新生、炎症反应及细胞凋亡的相关性。方法:选择在本院切除的瘢痕疙瘩标本作为研究的病理组,同期因外伤在成都市第三人民医院切除的正常皮肤组织作为研究的对照组。检测瘢痕疙瘩标本和正常皮肤组织中HIF-1α及血管新生分子、炎症反应细胞因子、细胞凋亡分子的表达量。结果:病理组瘢痕疙瘩中HIF-1α、VEGF165、Flt-1、Flk-1、Ang-1、Tie-2、PGE2、PGF2α、MIF、Livin、Survivin的mRNA表达量显著高于对照组正常皮肤组织,TSG-6、Caspase-3、Caspase-7、Caspase-9的mRNA表达量显著低于对照组正常皮肤组织;HIF-1α的mRNA表达量与VEGF165、Flt-1、Flk-1、Ang-1、Tie-2、PGE2、PGF2α、MIF、Livin、Survivin的mRNA表达量呈正相关,与TSG-6、Caspase-3、Caspase-7、Caspase-9的mRNA表达量呈负相关。结论:瘢痕疙瘩中HIF-1α呈高表达并且能够促进血管新生及炎症反应、抑制细胞凋亡。Objective:To study the expression of hypoxia inducible factor1α(HIF1α)in keloid and its correlation with angiogenesis,inflammatory response and apoptosis.Methods:Keloid samples removed in the Third People's Hospital of Chengdu between June2014and March2017were selected as the pathology group of the research,and normal skin tissues removed in the Third People's Hospital of Chengdu due to injury were selected as the control group of the research.The expression of HIF1α,angiogenesis molecules,inflammatory response cytokines and apoptosis molecules in keloid samples normal skin tissues were detected.Results:HIF1α,VEGF165,Flt1,Flk1,Ang1,Tie2,PGE2,PGF2α,MIF,Livin and Survivin mRNA expression in keloid of pathology group were significantly higher than those in normal skin tissue of control group while TSG6,Caspase3,Caspase7and Caspase9mRNA expression were significantly lower than those in normal skin tissue of control group;HIF1αmRNA expression was positively correlated with VEGF165,Flt1,Flk1,Ang1,Tie2,PGE2,PGF2α,MIF,Livin and Survivin mRNA expression,and negatively correlated with TSG6,Caspase3,Caspase7and Caspase9mRNA expression.Conclusion:HIF1αis highly expressed in keloid and can promote angiogenesis and inflammatory response and inhibit apoptosis.
