摘 要:目的观察柴胡疏肝散对功能性消化不良(FD)大鼠胃排空的促进作用,并探讨其作用机制。方法将24只健康成年SD大鼠随机均分为正常组、模型组、柴胡疏肝散组与多潘力酮组。除正常组外,其余各组均采用夹尾刺激法制备FD模型。各组均于造模第1天给予药物干预,柴胡疏肝散组、多潘立酮组分别给予0.63 g/mL柴胡疏肝散水煎剂1.5 mL、0.6 mg/mL多潘立酮水溶液1.5 mL灌胃,正常组、模型组均给予生理盐水1.5 mL灌胃,2次/d,共干预4周。末次给药后24 h,各组予以营养性半固体糊(1.5 mL/100 g体质量)灌胃,30 min后麻醉开腹,剪取完整胃,根据全胃重、空胃重计算胃排空率。用HE染色法观察胃窦部黏膜组织病理变化,用免疫组化法检测胃窦部黏膜组织中蛋白激酶R样内质网激酶(PERK)和磷酸化真核翻译起始因子2α(p-eIF2α)蛋白表达。结果与正常组比较,模型组大鼠胃排空率低(P<0.05);与模型组比较,柴胡疏肝散组、多潘立酮组胃排空率高(P均<0.05)。模型组、柴胡疏肝散组和多潘立酮组胃窦组织可见少量淋巴细胞及中性粒细胞浸润。与正常组比较,模型组大鼠胃窦组织内PERK、p-eIF2α蛋白表达高(P均<0.05);与模型组比较,柴胡疏肝散组、多潘立酮组胃窦组织内PERK、p-eIF2α蛋白表达低(P均<0.05)。结论柴胡疏肝散可能通过调控内质网应激信号通路PERK/eIF2α促进FD大鼠胃排空。ObjectiveTo observe the promotion of Chaihu Shugan powder on gastric emptying of functional dyspepsia(FD)rats and to investigate the underlying mechanism.MethodsTwenty four healthy adult SD rats were randomly divided into the normal group,the model group,the Chaihu Shugan powder group,and the domperidone group.The FD rat model was established by tail clamping in all groups except the normal group.All rats were medicated from the first day of modeling,rats in the Chaihu Shugan powder group and domperidone group were treated with0.63g/mL Chaihu Shugan powder decoction1.5mL and0.6mg/mL domperidone suspension1.5mL by gavage,respectively,and rats in the normal group and model group were administered normal saline1.5mL by gavage,all medications were implemented twice a day and for4weeks.At24hours after the last medication,all rats were fed with semisolid nutrient paste(1.5mL/100g weight)to get the whole stomach by laparotomy with anesthesia after30minutes,and then we calculated rates of gastric emptying on the basis of weights of total stomach and empty stomach.The histopathological changes of gastric antrum mucosa were observed by HE staining.The immunohistochemistry(IHC)technique was used to measure the protein kinase R like endoplasmic reticulum kinase(PERK)and phosphorylated eukaryotic translation initiation factor2αsignaling pathway(p eIF2α)protein expression levels of gastric antrum mucosa tissues in rats.ResultsCompared with the normal group,the rate of gastric emptying in the model group decreased(P<0.05);compared with the model group,the gastric emptying rates of the Chaihu Shugan powder group and domperidone group increased(both P<0.05).Cells in different tissue layers arranged well.A few lymphocytic and neutrophil infiltrated in antrum of rats in the model group,Chaihu Shugan powder group and domperidone group.Compared with the normal group,the expression of both PERK and p eIF2αincreased in the model group(both P<0.05);compared with the model group,the expression of PERK and p eIF2αdecreased in the Chai
