摘  要:目的:研究中心体扩增激酶STK15以及信号传导与转录激活因子3(STAT3)小干扰RNA(siRNA)在胃癌中的表达。方法:采用Real-time荧光定量PCR技术对7例胃癌新鲜组织和2例正常胃粘膜组织中STK15的mRNA表达水平进行检测。采用体外转染实验以及荷瘤裸鼠体内实验评价STAT3-siRNA的体内外抗肿瘤效果。结果:STK15在胃癌组织的表达量明显高于正常胃黏膜组织,差异有统计学意义(P<0.01)。另外,STAT3-siRNA能够显著降低胃癌细胞的体外增殖能力。结论:中心体扩增是细胞癌变过程的重要事件,中心体的异常扩增是细胞恶性克隆的关键环节。STAT3-siRNA显著降低胃癌细胞的体内外增殖能力和克隆形成能力。Objective:To study the expression of centrosome amplification kinase STK15and transcription factor3(S T A T3)small interfering RNA(siR N A)in gastric carcinoma.Methods:The expression of STK15mRN A in gastric tissue was detected by Real-time fluorescence quantitative PCR.In vitro and in vivo antitumor effects of STAT3-siRN A were evaluated by in vitro transfection experiments and tumor-bearing nude mice.Results:The expression of STK15in gastric cancer was1.326dz0.072,0.655dz0.053in normal gastric tissue,the difference was significant(P〈0.01).STAT3-siRN A significantly reduced the ability of gastric cancer cells to proliferate and clone formation ability in vitro.Conclusion:Central body amplification is an important event in the process of cell carcinogenesis.Abnormal amplification of central body is the key link of malignant cell cloning.STAT3-siRN A significantly reduced the ability of gastric cancer cells in vitro and in vivo and tumorigenic ability.Which may play a role by inhibiting the expression of PCNA.
