摘 要:目的探索DNA条形码技术在蝙蝠种类鉴定中的应用。方法本研究以舟山群岛地区3种常见蝙蝠标本为研究对象,提取基因组DNA,扩增CytB基因片段并测序,将测序结果进行BLAST比对分析;基于Kimura双参数模型计算22只蝙蝠CytB序列的种内种间遗传距离。结果遗传进化分析显示种间遗传距离显著大于种内遗传距离,通过NJ法构建系统发育树聚类分析,不同种类的蝙蝠位于不同的分支,同属的蝙蝠分支较不同属的蝙蝠分支较短,种内分支则更短,上述结果都与形态学鉴定结果一致。结论该研究建立的DNA条形码技术对于本研究中所涉及的蝙蝠种类具有较好的区分效果,可以作为一种有效的工具在蝙蝠鉴定中进行应用。Objective To study the application of DNA barcoding to identification of bats.Methods The CytB genes were amplified and sequenced from twenty-two bat samples collected across Zhoushan Islands.The intraspecific and interspecific genetic distances were calculated using the Kimura-2-parameters model.Results The interspecific genetic distances were significantly greater than the intraspecific ones.Rats of different species were in different branches.Branches in the same genus were shorter than those in different genera,and the branches of intraspecies were even shorter.These findings were consistent with the results of morphological identification.Conclusion This study shows that the species of bats can be well distinguished with CytB barcodes,and DNA barcoding can be used to effectively identify bat species.
