摘 要:目的研究铝暴露对大鼠肝脏葡萄糖转运蛋白4(GLUT4)表达的影响。方法将30只大鼠随机分成两组,即空白对照组(15只)和实验组(15只)。实验组予腹腔注射8mg/(kg·d^(-1))AlCl_3溶液(AlCl_3溶液用蒸馏水配制并高压灭菌)制成铝暴露大鼠模型,对照组给予腹腔注射等体积生理盐水(2ml/只),连续注射3d,休息1d再注射,全程共30d。于第10d、20d、30d取大鼠血清检测空腹血糖(FBG)、空腹胰岛素(FINS),采用稳态模式评估法(Homa)计算胰岛素抵抗指数(HOMA-IR),采用实时荧光定量PCR技术检测肝脏组织中GLUT4mRNA的表达,免疫组化法检测肝脏组织中GLUT4蛋白表达的情况。结果与对照组比较,实验组的FBG于第10d、20d高于对照组(P<0.05);随着染铝时间的延长,FINS表达呈下降趋势,于第10d、20d时明显高于对照组(P<0.05),第30d时反而低于对照组(P<0.05)。实验组的HOMA-IR随着暴露时间的延长,于第10d、20d时明显高于对照组(P<0.05),第30d时降低,与对照组比较差异无统计学意义。与对照组比较,实验组各时点的GLUT4mRNA及蛋白表达量明显降低(P<0.05)。结论铝暴露可降低大鼠肝脏GLUT4的表达水平,影响机体血糖代谢。Objective To study the influences of aluminum exposure on the expression of glucose transporter protein 4 in rat's liver. Methods Totally 30 rats were randomly divided into two groups:the control group(15 rats)and the experimental group(15 rats).The rat models of aluminum exposure of the experimental group were established by intraperitoneal injection with 8 mg/(kg·d^(-1))AlCl_3 solution(AlCl_3 solution was prepared with distilled water and sterilized by high pressure).Every rat model of the control group received intraperitoneal injection with same dosage of 2 ml NS.This experiment lasted for 30 days:continual injection for 3 days,rest 1 day,then repeated injection.On day 10,day 20 and day 30,the rat serum fasting blood-glucose(FBG)and fasting insulin(FINS)were measured and the insulin resistance index(Homa-IR)was calculated and analyzed with Homeostasis Model Assessment(Homa).The level of GLUT4 mRNA in liver was detected by Real-time Quantitative PCR technique,and the protein expression of GLUT4 was detected by Immunohistochemistry. Results The levels of FBG in experimental group rats were higher than those of the control group at day 10 and day 20(P <0.05).The level of FINS decreased with the prolongation of the rats exposure to aluminum.The rat FINS in the experimental group was significantly higher than those of the control group at day10 and day 20(P <0.05),but lower than that of the control group at day 30(P <0.05).HOMA-IR of the experimental group was significantly higher than that of the control group at day 10 and day 20(P <0.05)with the prolongation of exposure,but it decreased at day 30.The differences between experimental group and control group were not statistically significant.The expression levels of GLUT4 mRNA and protein in rat's liver in experimental group at every time point were significantly lower than those in control group(P <0.05).Conclusion Aluminum exposure can reduce the expression level of GLUT4 in rat's liver and affect blood glucose metabolism.
