摘  要:目的观察刺山柑总生物碱对系统性硬皮病(SSc)小鼠血管内皮生长因子(VEGF)、内皮素-1(ET-1)和可溶性血管细胞粘附分子-1(s VCAM-1)的影响,探讨其治疗SSc的作用机制。方法 90只BALB/c小鼠随机分为空白对照组、模型对照组、青霉胺组(125 mg·kg^(-1))及刺山柑总生物碱小(225 mg·kg^(-1))、中(450 mg·kg^(-1))、大(900 mg·kg^(-1))剂量组。除空白对照组外,其余5组小鼠背部注射盐酸博莱霉素建立SSc模型,成模后受试药组小鼠背部外敷刺山柑总生物碱乳膏,青霉胺组给予青霉胺灌胃,空白对照组、模型对照组背部外敷不含药基质,每天1次,连续60 d。末次给药后,酶联免疫吸附法检测小鼠皮肤组织VEGF及血清ET-1、sVCAM-1含量。结果与模型对照组比较,刺山柑总生物碱大剂量组SSc小鼠皮肤VEGF及血清ET-1水平降低(P<0.05或P<0.01),血清s VCAM-1含量无明显影响(P>0.05)。结论刺山柑总生物碱对SSc VEGF与ET-1异常表达具有一定调节作用。ObjectiveTo observe the effects of Capparis spinosa total alkaloid on vascular endothelial growth factor(VEGF),endothelin1(ET1)and soluble vascular cell adhesion molecule1(sVCAM1)levels in systemic sclerosis(SSc)mouse model and explore the therapeutic mechanism of Capparis spinosa total alkaloid for the treatment of SSc.MethodsA total of90BALB/c mice were randomly divided into blank control group,model control group,penicillamine(125mg·kg-1)group and Capparis spinosa total alkaloid low(225mg·kg-1),medium(450mg·kg-1)and high(900mg·kg-1)dose group.Except for the blank control group,SSc mouse model was established by daily subcutaneous injection of bleomycin in the back of the mice.After establishing model successfully,Capparis spinosa total alkaloid emulsifiable paste was externally applied with Capparis spinosa total alkaloid group,ground substance was externally applied to the mice in blank control group and model control groups,penicillamine was intragastrically administrated in the penicillamine group for60days,once daily.After the treatment,the content of VEGF in skin tissue and ET1,sVCAM1in serum were measured by ELISA.ResultsThe levels of VEGF and ET1were significantly decreased in Capparis spinosa total alkaloid high dose group as compared with model control group(P<0.05,P<0.01),but the content of sVCAM1wasn't influenced(P>0.05).
