摘 要:首次采用RAPD分子标记法研究内生真菌接种宿主红景天条件下,前者对后者DNA序列水平遗传性状的影响。结果显示,12条RAPD引物对ZPRs-R-11接种的红景天扩增出87条条带,多态性百分比为83.91%。聚类分析结果表明,内生真菌ZPRs-R-11接种的红景天可分为三个遗传分支,共培养时间越长,组培苗与内生真菌之间的遗传相似系数越大;红景天接种后,其基因流Nm为0.209 8。研究结果为内生真菌与宿主大花红景天共生条件下内生真菌对大花红景天的品质改良和研究提供参考。RAPD marker analysis was adopted firstly to investigate effects of genetic character Rhodiola crenulata inoculated with endophytic fungus ZPRs-R-11.The results showed that a total of87bands were amplified by12RAPD primers,and the percentage of polymorphism was83.91%in R.crenulata inoculated with ZPRs-R-11.Clustering analysis indicated that R.crenulata plants inoculated with ZPRs-R-11was clustered into three genetic branches.The similarity coefficient was higher when symbiosis between endophytic fungus and plant got more long time.The gene flows(Nm=0.2089<1)between fungus and host plant has lower value,which showed that genetic traits of Rhodiola plant could be affected deeply by endophytic fungus.The results in the present paper would be as scientific data for breed improvement and quality researching of Rhodiola plants.
