摘 要:目的:研究空间记忆不良的雌性SD大鼠的肠道菌群多样性和正常对照之间的差别和变化。方法 :应用Morris水迷宫实验方法对40只成年未孕雌性SD大鼠进行空间记忆学习能力的评估,筛选出空间记忆先天不良的SD雌性大鼠,并提取其与正常雌性大鼠的粪便DNA进行16S rRNA测序,并对测序结果进行比较和分析。结果:空间记忆不良的雌性SD大鼠在Morris水迷宫中的定位航行实验第5天找到平台的平均时间为(49.95±10.98)s,明显长于正常对照组(19.73±13.12)s(P=0.000)。空间记忆不良的雌性SD大鼠在空间探索试验实验中穿越目标象限次数(2.06±1.14)次,明显低于正常对照组(6.84±2.02)次(P<0.000 1)。空间记忆不良的雌性SD大鼠较正常对照的肠道菌群多样性减少,其中厚壁菌门丰度减少,拟杆菌门丰度明显增加,放线菌门丰度明显减少。相关性分析显示,放线菌门丰度和雌性SD大鼠空间记忆不良的程度呈显著负相关。结论:空间记忆不良的雌性SD大鼠肠道菌群构成较正常对照有明显变化,且这种变化可能参与了雌性SD大鼠生长发育过程中的脑部能量代谢和慢性炎症应激免疫应答过程。肠道菌群多样性研究可能成为筛选空间记忆不良的SD雌性大鼠的新的辅助评估方法和治疗干预的新靶点。To investigate the difference and change of gut microbiota diversity between the congenital impairedspatial memory female SD rats(CISMFS rats)and the normal ones.Methods:Using Morris watermaze to test the spatial memoryand learning of40SD female rats,CISMFS rats were screened out.The sequence information of the rats'feces were compared and analyzed based on16S rRNA gene sequence.Results:CISMFS rats spent significantly longer time[(49.95±10.98)s]to find the plat than the normal group[(19.73±13.12)s](P=0.000)during the fifth day of place navigation test.In the spatialprobe test,the CISMFS rats crossed the goal quadrant(2.06±1.14)less than the normal group(6.84±2.02)(P<0.0001).Theanalysis showed the gut microbiota diversity of the CISMFS rats decreased,the abundance of phylum Firmicutes decreased,the abundance of Bacteroides significantly increased,the abundance of phylum Actinobacteria decreased significantly compared to the normal group.Correlation analysis showed the abundance of phylum Actinobacteria were negatively correlatedwith the damage degree of spatial memory in CISMFS rats.Conclusions:The gut microbiota diversity of CISMFS rats was different from the normal rats which may contribute to the procedure of the brain energy metabolism and chronic inflammationwith immunity response.The diversity of gut microbiota could be a good supplement to evaluate the rodents'spatial memoryand be a new target of intervention.
