摘  要:探求由鸡爪苓菌丝体形成菌核的关键结构基因,为深入研究该珍稀药用真菌的生长发育、代谢调控以及优质种质资源的开发、利用与保护提供理论基础。从鸡爪苓菌核全长cDNA文库中随机筛选500个单独克隆进行单方向测序,选取3条全长的ESTs序列进行蛋白质功能预测,并进行生物学信息分析。结果表明:获得455条有效的EST序列,经聚类分析得到单基因簇共349条,文库冗余度为23%;参照拟南芥功能分类标准,将BLASTx比对结果中具有已知功能的78条单基因簇分成9类,分别是代谢、能量、转录、蛋白质合成、细胞结构、胞内运输、信号转导、抗性防御及未确定分类;仅有183条获得1条或1条以上GO术语,其中细胞组分包括7个部分,分子功能包括5个部分,生物学过程包括16个部分。Exploring the key genes concerned with formation of sclerotium form mycelia will provide a foundation for the growth and development of fungus,the elucidation of metabolic regulations,and the development and utilization of rare medicinal fungus.A total of500clones randomly selected from the sclerotium cDNA library were sequenced,and from which three full length ESTs were screened out and annotated their protein functions and biological information.The results showed that455high quality ESTs were obtained.Among these,369Tags were unigenes,and the redundancy of cDNA library accounted for23%.Referring to Arabidopsis thaliana function classification criteria,78known function unigenes by BLAST x were divided into9classes related with metabolism,energy,transcription,protein synthesis,cell structure,transport,signal transduction,defense and disease and the other unknown class,respectively.Only183unigenes got one or more than one GO term belonging to cell components consisted of7parts,molecular function included5parts,biochemical processes included16parts,respectively.
