摘  要:为研究东北刺人参不定根不同提取条件对酪氨酸酶活性抑制的影响,筛选最佳的提取工艺,采用多巴色素法,以东北刺人参生物反应器培养的不定根为材料,比较甲醇、乙醇和水提取物对酪氨酸酶抑制活性的影响;利用L9(34)正交试验法研究了提取时间、提取温度、提取次数和料液比的影响,优化不定根乙醇提取条件。结果表明:不定根乙醇提取物对酪氨酸酶的抑制率高于甲醇和水提物。正交试验结果发现,乙醇提取物的最佳提取工艺为提取时间1.5h,料液比1∶20,提取温度70℃,提取次数3次,在此条件下东北刺人参不定根提取物对酪氨酸酶活性的抑制率达到最高,为77.6%。To elucidate the effect of extraction conditions of Oplopanax elatus adventitious roots on inhibiting tyrosinase activity and to select an optimum extraction process,bioreactor cultured O.elatus adventitious roots as materials,the effects of methanol,ethanol and water extracts on inhibiting tyrosinase activity were determined.In addition,extraction temperature,extraction time,extraction times and material liquid ratio were clarified orthogonal experiments of L9(34),and the ethanol extraction condition was optimized.The results showed that the ethanol extract of adventitious roots had higher inhibitory rate of tyrosinase activity than methanol and water extracts.The orthogonal experiment revealed that the inhibitory rate of tyrosinase activity(77.6%)was the highest under the conditions that adventitious roots were extracted in80%ethanol for1.5h with1∶20of the material liquid ratio at70℃thrice.
