刺楸愈伤组织诱导及其再分化的研究

摘　　要：试验以刺楸叶片为外植体,研究了影响愈伤组织诱导和再生的因素。结果表明:培养基种类对诱导愈伤组织有影响,在MS基本培养基中愈伤组织诱导率最高,达89.3%。愈伤组织在含KT(激动素)2.0 mg/L、NAA(萘乙酸)0.10mg/L的MS培养基中,再分化效果最好。蔗糖作为糖源时,愈伤组织的再分化效果显著高于其他碳源。不定芽的分化随着培养时间的增加呈先增后减的趋势,在培养40d时芽数最多,产生12.4个不定芽,分化率达93.0%,芽平均长1.2cm。The leaves of Kolopanax septemlobus as explants,the effects of the factors on callus induction and its rediffererntitation was determined.The results showed that the types of culture medium had an effect on the induction of callus,and the highest induction rate of89.3%was obaited on MS medium,whereas the best redifferentiation effect was obtained on the medium of MS adding2.0mg/L of kinetin and0.10mg/L of naphthylacetic acid.When the sucrose was used as carbon source,the effect of callus redifferentiation was significantly higher than the case with other carbon sources.The growth of buds increased firstly and then decreased with the elongation of culture time,and the greatest number of buds with12.4buds was produced when cultured for40days with the differentiation rate of93.0%,and the average length of buds was1.2cm.
