三种分色方法制作旋毛虫肌幼虫囊包染色标本的效果观察

摘　　要：[目的]观察三种分色方法制作旋毛虫肌幼虫囊包染色标本的效果。[方法]分色方法 1:染色片经体积分数为50%、70%、80%、95%的乙醇脱水,然后入体积分数为2%的盐酸酒精中分色,再经体积分数为100%的乙醇脱水。分色方法 2:染色片入体积分数为2%的盐酸酒精第一次分色,入体积分数为50%、70%、80%、95%的乙醇脱水,再入体积分数为2%的盐酸酒精第二次分色,然后入体积分数为100%的乙醇脱水。分色方法 3:染色片入体积分数为2%的盐酸酒精分色,入体积分数为50%、70%、80%、95%、100%的乙醇脱水。[结果]三种分色方法制作的囊包染色标本,囊包轮廓清晰,囊包、囊内幼虫和周围肌纤维色泽差别较大,结构典型,易于观察。[结论]制作旋毛虫肌幼虫囊包染色标本,分色方法 1的先脱水后分色,分色方法 2为脱水过程伴随两次分色,分色方法 3为先分色后脱水,均可得到较好的分色效果,关键是分色时间要掌握恰当。〔Objective〕To observe the effect of three color separation methods for preparing staining specimen of Trichinella Spiralis encapsulated Larvae.〔Methods〕Color separation method1:the dyeing pieces were dehydrated with50%,70%,80%,95%ethanol,then colors were separated in2%hydrochloric acidic alcohol,and dehydrated with100%ethanol;Color separation method2:the dyeing pieces were conducted the first color separation in2%hydrochloric acidic alcohol,dehydrated in50%,70%,80%,95%ethanol,the second color separation was conducted in2%hydrochloric acidic alcohol once more,and then dehydrated in100%ethanol;Color separation method3:the dyeing pieces were conducted the first color separation in2%hydrochloric acidic alcohol,dehydrated in50%,70%,80%,95%,100%ethanol.〔Results〕Encapsulated Larvae dyeing samples were prepared by3color separation methods,the contour of the capsule was clear,there existed the color difference in the capsule,the larva of the capsule and the surrounding muscle fibers.the structure was typical and it is easy to observe.〔Conclusion〕In make dyeing specimens of Trichinella Spiralis encapsulated Larvae,the ideal color separation effect can be received with the following3color separation methods,that is,color separation method1(dehydration first and color separation second),color separation method2(dehydration companied by two times of color separation),color separation method3(color separation first and dehydration second).The key is to decide the appropriate color separation time.
