摘 要:目的探讨丙烯酰胺对小鼠卵母细胞成熟及发育潜能的影响。方法选择成年雌性小鼠分别给予生理盐水及丙烯酰胺5mg/kg(低剂量组)、10mg/kg(中剂量组)、20mg/kg(高剂量组),连续给药7d,统计卵母细胞成熟率、孤雌激活率;观察卵母细胞纺锤体和染色体形态;分析卵母细胞外卵丘颗粒细胞的凋亡。结果 (1)丙烯酰胺低、中、高剂量组卵母细胞的体外成熟率分别为(88.5±1.5)%、(79.5±1.7)%、(76.5±1.8)%,均低于对照组的(92.5±1.2)%,但只有中、高剂量组与对照组有统计学差异(P<0.05);(2)低剂量组卵母细胞的孤雌激活率[(78.5±2.6)%]与对照组[(81.5±1.2)%]相仿(P>0.05),而中、高剂量组卵母细胞的孤雌激活率[分别是(61.6±3.2)%、(40.3±5.6)%]均显著低于对照组(P<0.01);(3)丙烯酰胺处理组卵母细胞纺锤体形态不正常,呈现筒形和染色体排列不规则,散乱;(4)丙烯酰胺促进了颗粒细胞的凋亡。结论小鼠卵母细胞在丙烯酰胺处理后细胞成熟延迟,发育潜能降低。Objective:To explore the effect of acrylamide on oocyte maturation and developmental potential in mice Methods:The adult female mice were treated with different doses(5,10and20mg/kg)of acrylamide uninterruptedly for7days Then the mice were sacrificed to collect the oocytes The percentage of oocytes maturation and parthenogenetic activation rate were counted,and the spindle and chromosome morphology of oocyte and the apoptosis of cumulus cells were observed and analyzed Results:The in vitro maturation(IVM)rate of oocytes in low,medium and high dose acrylamide groups was(88.5±1.5)%,(79.5±1.7)%and(76.5±1.8)%respectively,which was lower than that in the control group(92.5±1.2%),but only the IVM rate in the medium and high dose groups was significantly different with the control group(P<0.05)The parthenogenetic activation rate of oocytes in the low dose group(78.5±2.6%)was not significantly different with the control group(81.5±1.2%)(P>0.05),but which in middle(61.6±3.2%)and high dose groups(40.3±5.6%)was significantly lower than that in control group(P<0.01)The oocytes displayed barrel spindle and the chromosomes became scattered and irregular in the acrylamide treated groups Acrylamide increased apotosis in the cumulus cells Conclusions:The maturation of mouse oocytes was delayed and the developmental potential decreased after treatment with acrylamide
