摘  要:以非洲菊(Gerbera jamesonii)嫩叶为材料,研究了不同外植体部位、暗培养条件、培养基激素等因素对非洲菊愈伤组织诱导和增殖的影响。结果显示,非洲菊叶片边缘切口出愈伤速度快于叶柄伤口;带叶基叶柄出愈伤率最高,诱导培养15 d出愈伤率为84%以上;叶柄上诱导的愈伤团生长最快,15 d愈伤生长量在82%以上。愈伤组织诱导期间使用暗培养能降低外植体死亡率,并一定程度上提高愈伤生长量,但显著降低了外植体诱导出愈伤率和愈伤质量。愈伤增殖培养最适培养基为MS+TDZ 1.5 mg/L+2,4-D0.30 mg/L,20 d内愈伤增殖量达到76%以上,有效降低了愈伤组织褐化死亡率,且愈伤组织状态良好。The effects of different explant parts,darkness plans and honnones on callus induction and proliferation were studied with unlolded leaf materials in Gerbera jamesonii.The results showed that the callus induction of blade explants was faster than petiole explants.Callus induction rate of winged petiole in15days was higher than84%,while unwinged petiole explants had a highest callus growth rate of over82%.Using darkness in callus induction had adverse effects on callus induction and quality,though a good for callus volume growth.MS+TDZ1.5m g/L+2,4-D0.30m g/L was the best medium for callus proliferation,with a callus proliferation rate higher than76%in20days,and good callus tissues with lower death rate from browning as well.
