Protective Effects of Tea Saponins Derived from Gynostemmapenta phyllum Against High Dietary Choline-Induced Endothelial Injury in Mice

摘  要:为了探索质量分数为3%高胆碱饮食对小鼠血管内皮功能的损伤程度以及绞股蓝茶皂苷潜在的心血管保护作用。连续8周饮食3%高胆碱后,小鼠血清中一氧化氮合酶(eNOS)、一氧化氮(NO)和前列腺素(PGI2)水平均显著降低(p<0.01),内皮素-1(ET-1)和血栓素(TXA2)水平均显著升高(p<0.01)。此外,小鼠胸主动脉血管组织的苏木精伊红切片也表明3%高胆碱饮食8周可导致典型的血管内皮损伤。然而,经连续8周的绞股蓝茶皂苷的营养干预后,血清生化指标及血管组织病理切片均得到显著缓解(p<0.05,p<0.01)。结果表明,高胆碱饮食可诱导小鼠血管内皮功能异常,而绞股蓝茶皂苷可有效干预高胆碱饮食导致的血管内皮损伤,是一种潜在的预防心血管损伤的营养物质。This studyexplored the vascular endothelialdamages in the mice fed high-choline diet(3%),andthe possible vasoprotective effects of tea saponins in Gynostemma pentaph/Hum(TSGP).After mice fed3%high choline water for continuous8weeks,the serumlevels of eNOS,NO and PGI2significantly reduced(p<0.01),while the serumlevels of ET-1and TXA2remarkably increased(p<0.01).Furthermore,the further histopathological observation by conventional H&Ealsosuggested thatvascular endothelialdamages vascularhappened in thoracic aorta after feeding3%high-choline diet for8weeks.However,the TSGP remarkably ameliorated the aboveserum biochemical indexsand the histological section after treating with TSGP for8weeks(p<0.05,p<0.01).This results showing that high intake of dietary choline may inducevascular endothelial dysfunction,while TSGPexerted a significant protective effect on the choline-induced vascular endothelialinjury and isa kind of potential nutrients to prevent cardiovascular damage.
