蠲痹历节清酊对大鼠急性痛风性关节炎模型滑膜组织中ICAM-1、NF-κB表达的影响  

Effects of Juanbi Lijieqing Tincture on ICAM-1, NF-κB Expression of Synovial Tissue in Model Rats with Acute Gouty Arthritis

摘　　要：目的初步探讨蠲痹历节清酊对大鼠急性痛风性关节炎模型的作用机制。方法选用SPF级健康雄性SD大鼠40只,分成蠲痹历节清酊组、扶他林软膏组、模型组、空白对照组,每组10只。对前三组采用踝关节腔注射尿酸钠溶液造成急性痛风性关节炎模型,并予以相应干预。观察各组大鼠造模后一般状态;用缚线法检测大鼠踝关节肿胀度;光镜下观察踝关节滑膜的病理变化;采用免疫组化染色检测滑膜组织中细胞间粘附分子-1(ICAM-1)和核转录因子(NF-κB)含量。结果与模型组比较,扶他林软膏组、蠲痹历节清酊组肿胀度降低(P<0.05);大鼠踝关节滑膜光镜观察:空白对照组滑膜组织结构整齐、层次分明。模型组滑膜组织结构损坏、层次混乱,组织充血、水肿,大量炎性细胞浸润、成纤维细胞增生,少量巨噬细胞浸润;扶他林软膏组、蠲痹历节清酊组与模型组比较改善明显;大鼠踝关节滑膜组织中,空白对照组与模型组ICAM-1、NF-κB表达比较差异有统计学意义(P<0.05)。扶他林软膏组、蠲痹历节清酊组与模型组ICAM-1、NF-κB表达比较差异有统计学意义(P<0.05)。结论蠲痹历节清酊具有缓解肿胀、改善滑膜炎症、抑制关节滑膜中ICAM-1、NF-κB表达的作用。Objective To explore the effect mechanism of Juanbi Lijieqing tincture on the model rats with acute gouty arthritis.Methods The40SPF level healthy male SD rats were divided into Juanbi Lijieqing tincture group,Futaling ointment group,model group,blank control group,10rats in each group.The former three groups were established acute gouty arthritis model,and intervened by Futalin ointment and Juanbi Lijieqing tincture.Rats were observed after modeled in general condition,then compared.The ankle swelling of rats after drug intervention was detected by the line method;the ankle synovial pathological changes were observed under light microscope;the ankle joint synovial tissue intercellular adhesion molecule1(ICAM-1),and nuclear-transcription factor-κB(NF-κB)in rats were determined by immunohistochemical staining method.Results The swelling degree of Futalin ointment group and Juanbi Lijieqing tincture group,compared with the control group(P<0.05).The synovial tissue structure in the blank group was neat and clear under the light microscope,while the synovial tissue structure in model group was disorder,synovial hyperemia,edema,and a large number of infiltration of inflammatory cells and proliferation of fibroblast,a small amount macrophage infiltration were also obseved in the model group;Futalin ointment group and Juanbi Lijieqing tincture group showed obvious improvement,compared with the model group.The expression of ICAM-1and NF-κB between the blank control group,Futalin ointment group and Juanbi Lijieqing tincture group with the model group were significantly different(P<0.05).Conclusion Juanbi Lijieqing tincture shows the effects of relieving swelling,improving synovial inflammation,inhibiting the expression of ICAM-1and NF-kB in the synovial membrane of the joint.
