摘 要:目的探讨[Gly14]-Humanin(HNG)预处理对局灶性脑缺血再灌注损伤大鼠氧化应激及神经细胞凋亡的影响。方法采用改良的Zea-longa法建立大鼠大脑中动脉阻塞再灌注损伤模型。96只雄性SD大鼠随机分为假手术组、模型组、生理盐水组和HNG组。HNG组大鼠造模前3天给予5μL HNG(100 nmol/m L),股静脉注射,连续3天,每天1次;假手术组与生理盐水组给予5μL生理盐水,缺血3 h再灌注24 h后,采用Zea-longa 5分法对各组大鼠进行神经功能缺损评分(NDS);检测脑组织中超氧化物歧化酶(SOD)活性、谷胱甘肽(GSH)及丙二醛(MDA)含量;原位末端标记法观察脑组织细胞凋亡情况。结果假手术组、模型组、生理盐水组和HNG组NDS的中位数(四分位数间距)分别为0(0)、1.5(1)、3(1)和3(1)分,假手术组NDS低于其他3组,HNG组NDS低于模型组和生理盐水组(均P<0.05)。与假手术组大鼠比较,模型组、生理盐水组及HNG组大鼠脑组织SOD活性[分别为(11.65±1.66)、(12.15±1.56)和(19.43±1.47)U/m L]及GSH水平[分别为(8.84±1.23)、(7.51±1.16)和(11.17±1.67)mg/L]均降低,MDA水平[分别为(42.61±1.79)、(40.33±1.24)和(35.39±1.29)nmol/m L]升高(P<0.05);HNG组SOD活性及GSH水平高于模型组及生理盐水组,而MDA水平低于模型组及生理盐水组(均P<0.05)。与假手术组[(0.13±0.01)%]比较,模型组、生理盐水组及HNG组凋亡细胞的百分率[分别为(0.59±0.07)%、(0.51±0.08)%和(0.22±0.03)%]均增多(P<0.05),HNG组大鼠凋亡细胞的百分率低于模型组和生理盐水组(均P<0.05)。结论 HNG可能通过增加脑组织SOD活性及GSH水平,抵抗缺血再灌注应激损伤,减少神经细胞凋亡,从而减轻神经功能缺损。Objective To study the effect of[Gly14]-Humanin on oxidative stress and neuron apoptosis after focal cerebral ischemia reperfusion injury in rats.Methods A rat model of acute middle cerebral artery occlusion(MCAO)and reperfusion was established by suture embolism.Ninety-six healthy male Sprague-Dawley rats were randomly divided into sham-operation group,model group,normal saline group and HNG group.And5滋L HNG(100nmol/L)in the rats of the HNG group,normal saline in the rats of the sham-operation group and normal saline group were given to rats3days before operation respectively(iv,three times,qd).After cerebral ischemia3h and24h of reperfusion,neurological deficit score(NDS)were performed for each group,and the activity of sod,levels of GSH and MDA in cerebral tissue were detected.TUNEL staining were used to detect neuronal apoptosis.Results Median(Interquartile range)of NDS was0(0)in sham-operation group,1.5(1)in model group,3(1)in normal saline group and3(1)in HNG group,respectively,and the NDS of sham-operation group was significantly lower than the other three groups.The NDS of HNG group was significantly lower than the model group and normal saline group(all P<0.05).Compared with sham-operation group,the activity of SOD of model group,normal saline group and HNG group rats[(11.65±1.66),(12.15±1.56)and(19.43±1.47)U/mL]and levels of GSH[(8.84±1.23),(7.51±1.16)and(11.17±1.67)mg/L]were decreased,and levels of MDA[(42.61±1.79),(40.33±1.24)and(35.39±1.29)nmol/mL]were increased(P<0.05).The activity of SOD and levels of GSH of HNG group were significantly higher than model group and normal saline group,but the levels of MDA was significantly lower than model group and normal saline group(all P<0.05).Compared with sham-operation group[(0.13±0.01)%],percentage of neuronal apoptosis of model group,normal saline group and HNG group rats[(0.59±0.07)%,(0.51±0.08)%and(0.22±0.03)%,respectively]were increased(P<0.05),and percentage of neuronal apoptosis of HNG group was significantly lower than model group and
