经抗菌素修饰的Brindley电极体外抗菌活性的可行性研究

摘  要:目的:目前Brindley电极植入后感染是其失败的主要原因之一,常需取出电极才能控制感染,为解决这一难题,笔者设计了经抗菌素修饰的Brindley电极,以初步探讨其抗感染能力。方法:根据制作方法不同分成两组:试验组按Bayston R介绍的浸渍法制作,选用利福平-克林霉素-三氯生作为抗菌素;对照组按传统方法用硅胶包裹制作,选用夫西地酸钠作为抗菌素。通过观察两组对大肠杆菌、表皮葡萄球菌的抑菌效应(抑菌圈的大小),来判定其抗菌活性。结果:当以大肠杆菌作为实验对象时,在第1个10天,试验组抑菌圈大小为(22.21±0.75)mm,对照组为(6.10±2.60)mm,两组的抑菌效应有显著性差异(P<0.05);在接下来的20天,对照组未显示出抑菌效应,两组不具有可比性;当以表皮葡萄球菌作为实验对象时,在3个时间段,试验组与对照组的抑菌效应比较,差异有统计学意义(P<0.05)。结论:经过利福平-克林霉素-三氯生三种抗菌素修饰的Bringdly电极具有持久的抗菌活性,且优于传统方法。Objective Currently infection after inplantation of Brindley electrode was a major reason for its failure,and often remove the electrode to control the infection,in order to solve this problem,we designed a modified Brindley electrode by antibiotics to explore its antimicrobial activity.Method According to different methods divided into two groups:Experimental group,using impregnation method by Bayston R,selection of rifampicin-clindamcin-triclosan as antibiotics;Control group,using in the traditional method wrapped silicone,the choice of fusidic sodium as the antibiotics.By observed inhibitory effect(inhibition zone size)respectively for E.coli,Staphylococcus epidermidis,to determine its antibacterial activity.Results Experriment showed that when E.coli as an experimental object,in the first ten days,inhibition zone size for the experimental group(22.21±0.75)mm,the control group(6.10±2.60)mm,there was significant difference between experimental group and control group in antibacterial effect(P<0.05);in the next20days,the control group did not show the inhibition effect,so the two groups were not comparable.when staphylococcus epidermidis as the test object,in three time periods,there was significant difference between experimental group and control group in antibacterial effect(P<0.05).Conclusion The modified Bringdly electrode by rifampicin-clindamcin-triclosan has long-lasting antimicrobial activity,and better than traditional methods..
