门氏定喘汤对吸烟哮喘大鼠IL-17A、IL-6、TNF-α的影响

摘　　要：目的:研究门氏定喘汤对吸烟哮喘大鼠IL-17A、IL-6、TNF-α的影响,为中药治疗支气管哮喘提供实验依据。方法:用OVA、Al(OH)3混悬液致敏大鼠及被动吸烟装置制作吸烟哮喘模型。A组为正常对照组,B组为哮喘吸烟模型组,C组为模型+地米组,D组每次雾化吸入激发前0.5h给予门氏定喘汤灌胃,E组每次雾化吸入激发前0.5h给予腹腔注射地米及门氏定喘汤灌胃,最后一次激发24h后将大鼠使用10%水合氯醛麻醉,收集肺泡灌洗液(BALF),使用ELISA法检测IL-17A、IL-6、TNF-α含量及灌洗液中细胞分类。结果:比较各组大鼠BALF中中性粒细胞及嗜酸性粒细胞数量,D组与C组无显著性差异,较B组有所降低;E组优于D组,有显著差异。各组大鼠IL-17A、TNF-α含量比较显示:E组、C组IL-17A、IL-6、TNT-α低于β组,E组IL-17A、TNT-α低于D组,P>0.05,有显著性差异。结论:中药门氏定喘汤联合地米治疗吸烟哮喘大鼠优于地米及单纯中药,且该中药能降低吸烟哮喘大鼠IL-17A、TNF-α含量,改善气道的炎性反应。Objective:To study the effect of the Men's Dingchuan decoction on IL-17A,IL-6,TNF-αin asthmatic rats and to provide experimental evidence for the treatment of bronchial asthma with TCM medicine.Methods:OVA and Al(OH)3suspensions were used to sensitize rats,and a passive smoking device was used to make asthma and smoking models.Group A was the normal control group,group B was the model group,group C was the dexame thasone group,group D was given the Men's Dingchuan decoction before half an hour in inhalation,and group E was given dexamethasone plus the Men's Dingchuan decoction before half an hour in inhalation.The rats were anesthetized with10%chloral hydrate and the alveolar lavage fluid was collected.The levels of IL-17A,IL-6and TNF-αand cell sorting in lavage fluid were detected by ELISA.Results:There was no significant difference in the number of neutrophils and eosinophils between group D and group C,and the number was more decreased than group B.And that in group E was better than group D,with a significant difference.The levels of IL-17A,IL-6and TNF-αin group E were lower than group B,and IL-17A,TNT-αin group E were lower than group D,with significant differences.Conclusion:The Men's Dingchuan decoction plus hormone was effective on asthmatic rats,by reducing IL-17A,TNF-α,and relieving the inflammatory response of airway.
