摘  要:目的探讨A型钾通道是否参与臂旁外侧核(LPB)神经元温度敏感性的形成。方法采用红外可视脑片膜片钳胞外记录技术,观察A型钾通道阻断剂4-氨基吡啶(4-AP)对LPB热敏、中斜率和低斜率温度不敏感神经元放电频率、放电幅度和温敏系数的影响。结果灌流4-AP引起热敏和低斜率温度不敏感神经元的放电频率明显加快(P<0.05);中斜率温度不敏感神经元放电频率无明显变化(P>0.05);4-AP对三类神经元放电幅度均无明显影响(P>0.05);灌流4-AP后,热敏神经元温敏系数有降低趋势;中斜率温度不敏感神经元温敏系数无明显变化(P>0.05);低斜率温度不敏感神经元温敏系数明显增加(P<0.05)。结论 4-AP对LPB不同类型神经元放电活动影响不同,A型钾通道可能参与了LPB神经元温度敏感性的形成。Objective To investigate whether A-type potassium channels are involved in the formation of neuronal thermosensitivity of lateral parabrachial(LPB)neurons.Methods The loose-seal cell-attached patch clamp recordings were used to observe the effect of the A-type potassium channel blocker 4-aminopyridine(4-AP)on the firing frequency,discharge amplitude and thermal coefficients of thermosensitive neurons and both medium-slope and low-slope temperature-insensitive neurons.Results The perfusion of 4-AP made the firing frequency of the thermosensitive and low-slope temperature-insensitive neurons accelerated significantly(P<0.05),while the firing frequency of medium-slope temperature-insensitive neurons was not changed significantly(P>0.05).4-AP did not significantly affect the discharge amplitude of the three types of LPB neurons(P>0.05).4-AP decreased the thermal coefficients of thermosensitive neurons and significantly increased those of low-slope temperature-insensitive neurons(P<0.05),while it had no significant effect on the thermal coefficients of medium-slope temperature-insensitive neurons(P>0.05).Conclusion 4-AP has different effect on the firing activity of three types of LPB neurons,and A-type potassium channels may be involved in the formation of neuronal thermosensitivity of LPB neurons.
