摘  要:目的以线粒体为研究对象探讨蛇床子总香豆素对心肌梗死大鼠心脏的影响。方法采用冠脉结扎法制作大鼠心肌梗死模型,给予蛇床子总香豆素8周后,H-E染色检测大鼠心肌梗死程度,Power Lab系统检测心脏功能,透射电镜观察心肌线粒体排列和形态,Western Blot技术检测线粒体形态相关蛋白。结果与模型组相比,给予心肌梗死大鼠蛇床子总香豆素能够显著降低其心肌纤维化,改善心脏功能,减少心肌线粒体碎片,下调Drp1并上调OPA1的表达,改善线粒体形态。结论蛇床子总香豆素可通过改善线粒体形态降低心肌梗死大鼠的心肌损伤。Objective To explore the protective effect of total coumarines from the fruits of Cnidium monnieri on mitochondrial dynamics in rat myocardial infarction model through the regulation of mitochondrial morphology.Methods Left anterior descending coronary artery ligation of rats were performed to induce the myocardial infarction model.H-E staining was ligated to measure the infarct area and cardiac fibrosis.Powerlab system was used to detect cardiac hemodynamic parameters.The arrangement and morphology of mitochondria on rat heart tissue were observed by transmission electron microscopy and mitochondrial morphology related proteins were measured by using Western Blot.Results Compared with the model group,the rats treated with total coumarines exhibited a significant decrease in cardiac infarction area and cardiac fibrosis,and the hemodynamic parameters of coumarines group were also improved.The cardiac mitochondrial fragments in coumarines group were obviously less than that in model group;the expressions of Drp1and OPA1were reversed in coumarines group compared with model group.Conclusion Total coumarines from the fruits of Cnidium monnieri has a protective effect on myocardial infarction in rats,which is associated with the improved mitochondrial morphology.
