摘 要:目的:观察莪术油对子宫内膜异位症模型大鼠异位内膜组织中基质细胞衍生因子-1(SDF-1)及其趋化因子受体4(CXCR4)表达的影响。方法:采用自体移植法建立非动情期SD大鼠子宫内膜异位症模型,将造模成功的32只大鼠随机分为模型组、莪术油(低、中、高剂量)组,每组8只,另设8只健康大鼠作为空白组;连续给药28天后观察异位内膜生长情况,切取在位及异位内膜组织,行PCR法检测大鼠在位、异位内膜组织及正常在位子宫内膜组织SDF-1和CXCR4的mRNA表达情况,对比莪术油各剂量组与模型组异位内膜组织中SDF-1 mRNA,CXCR4 mRNA的表达差异。结果:大鼠异位内膜组织的SDF-1 mRNA和CXCR4 mRNA高度表达,与在位内膜组织及正常子宫内膜组织比较差异有统计学意义(P<0.05);莪术油各剂量组异位内膜组织SDF-1mRNA表达均有不同程度下降,且各剂量组间比较差异有统计学意义(P<0.05);莪术油各剂量组CXCR4 mRNA表达亦有下降趋势,各剂量组间比较差异有统计学意义(P<0.05);莪术油各剂量组SDF-1 mRNA和CXCR4 mRNA的表达均显著低于模型组。结论:SDF-1/CXCR4轴可能参与了子宫内膜异位症的发展过程;莪术油可明显降低异位内膜组织中SDF-1、CXCR4表达,对子宫内膜异位症起一定的治疗作用。Objective:To investigate the influence of curcuma zedoary oil on stromal cell derived factor-1(SDF-1)and its receptor CXC chemokine receptor(CXCR4)expressions in endometriosis(EM)rats model.Methods:The EM rat models were established by self-endometrium transplantation during non-estrus period and randomly assigned into the model group,the high-dose group,the middle-dose group and the low-dose group(eight rats in each group),eight healthy rats were taken as the blank group.After28days of successive administration,the growth of ectopic endometria was observed;the eutopic and ectopic endometria were removed in preparation for PCR method in test of the expression of SDF-1and CXCR4mRNA,to obtain the difference between the treatment group and the control group.Results:SDF-1and CXCR4mRNA were highly expressed in ectopic endometrial tissue,there is astatistical difference in comparison with eutopic endometria and normal endometria tissue(P<0.05);the expressions of SDF-1mRNA in ectopic endometrial tissue declined in curcuma zedoary oil groups(low,middle and high doses)to different degrees,and the differences had statistical meaning among different doses groups(P<0.05);the expressions of CXCR4mRNA showed the tendency of declining in curcuma zedoary oil groups(low,middle and high doses),and the difference had statistical meaning among different doses groups(P<0.05);curcuma zedoary oil groups(low,middle and high doses)were lower than the model group in the expressions of SDF-1mRNA and CXCR4mRNA.Conclusion:SDF-1/CXCR4axis might participate in the developmental process of endometriosis;curcuma zedoary oil could lower the expressions of SDF-1and CXCR4in ectopic endometrial tissue and it shows certain therapeutic effects in treating EM.
