摘 要:目的探讨七叶皂苷钠对油酸诱导的急性肺损伤纤维化大鼠的干预作用。方法实验动物随机分成正常对照组、模型组、七叶皂苷钠干预组,制备大鼠油酸型急性肺损伤纤维化大鼠模型,七叶皂苷钠干预组大鼠静脉注射七叶皂苷钠注射液4 mg/kg,连续14 d,分别于末次给药后第1天和第14天(停药1 d和14 d)各组随机处死11只大鼠取材,观察模型大鼠体质量的变化,测定动脉血氧分压(Pa O2)和氧合指数(Pa O2/Fi O2)、肺指数及湿干质量比、血清IL-1β、TNF-α、PCⅢ、TGF-β1水平,进行肺组织病理学检查。结果各组动物体质量、肺指数及湿干质量比差异不明显,与模型组相比,七叶皂苷钠干预组Pa O2和Pa O2/Fi O2明显升高(P<0.05),IL-1β、TNF-α、PCⅢ、TGF-β1水平显著降低(P<0.05),肺组织病理学损伤明显减轻,肺损伤组织学半定量评分(IQA)分值显著降低(P<0.01)。结论七叶皂苷钠能降低急性肺损伤大鼠模型炎症因子水平,对肺纤维化具有一定的干预作用。Objective To explore the intervening effect of sodium aescinate on pulmonary fibrosis in ALI rats.Methods The rats were randomly divided into the normal group,the model group and sodium aescinate group.The rat model of acute lung injury was induced by administration of oleic acid.Sodium aescinate group rats were intravenously injected of sodium aescinate for14days.The11rats were sacrificed at1st and14th day after last administration.The indexs of weight,arterial partial pressure of oxygen(PaO2),oxygenation index(PaO2/FiO2),lung index and wet/dry ratio,IL-1β,TNF-α,PCⅢ,TGF-β1and the lung histopathology in rats were observed.Results Compared with the model group,there was no difference in rat weight,lung index and wet/dry ratio.After treatment with sodium aescinate,the levels of PaO2and PaO2/FiO2were increased.Meantime,the levels of IL-1β,TNF-α,PCⅢ,TGF-β1and IQA were reduced,and the lung histopathology damage was lighten.Conclusion Sodium aescinate can reduce the levels of inflammatory factors,which shows a preventive effect on pulmonary fibrosis to ALI rats.
