摘 要:目的:探究不同种大黄中土大黄苷检测方法。方法:采取HPLC和TLC测定不同种大黄中土大黄苷的含量。色谱柱C18柱(Aglient TC-C18analytical,4.6 mm×250 mm,5μm),流动相乙腈-水(27∶75),柱温30.0℃,检测波长323 nm,流速0.80 m L/min,用DAD检测器对对照品和供试品溶液进行光谱扫描。结果:药用大黄中没有出现土大黄苷特征性色谱峰,河套大黄和华北大黄均出现土大黄苷特征性色谱峰(土大黄苷仅出现于伪品大黄中);土大黄苷对照品和供试品中出现了相同的土大黄苷特征性色谱峰,并且两色谱峰在DAD检测器下扫描得到的光谱图一致。结论:采取HPLC法及TLC法鉴别大黄药材有简单、快捷、准确等优势,可作为检测不同种大黄中土大黄苷的推荐方法。Objective To explore the detection methods of rhubarb glycosides in different rhubarb.Methods HPLC(high performance liquid chromatography)and TLC(thin layer chromatography)determination of the rehmannia glycosides is different rhubarb.The mobile phase consisted of acetonitrile-water(27∶75)with a column temperature of 30.0℃and a detection wavelength of 323 nm at a flow rate of 0.80 mL/min on a C 18 column(Aglient TC-C 18 analytical,4.6 mm×250 mm,5μm)Product and the test solution for spectral scanning.Results Rhubarb Medicinal Rhubarb did not appear characteristic chromatographic peaks,Rhododendron and Datong Rhubarb are featured Indoloine peak characteristic chromatin(emodin appears only in the pseudo-rhubarb).Soil emodin glycosides reference substance and the test sample appeared the same euphorbia glycosides characteristic peaks,and the two peaks were scanned under the DAD detector obtained spectrogram consistent.Conclusion The HPLC method and TLC method to identify Rhubarb is simple,fast and accurate and can be used as a method to detect the different Rhubarb.
