牛蒡苷元联合帕金森药物对鱼藤酮诱导的PC12细胞损伤的保护作用

摘　　要：目的:研究牛蒡苷元对鱼藤酮诱导的PC12细胞损伤的保护作用并探讨其联合阳性药(甲磺酸溴隐亭或盐酸苯海索)是否具有协同作用。方法:通过不同浓度牛蒡苷元和阳性药处理鱼藤酮诱导的PC12细胞,采用MTT法测定细胞存活率,评价牛蒡苷元和阳性药对PD细胞模型的保护作用,并通过Compu Syn软件计算两药的联合药物指数(combination index,CI)。结果:牛蒡苷元、甲磺酸溴隐亭和盐酸苯海索均可显著提高鱼藤酮损伤PC12细胞的存活率(P≤0.01),周-特氏联合指数法表明,牛蒡苷元与甲磺酸溴隐亭(比例1∶4和1∶2)、与盐酸苯海索(比例1∶1和2∶1)联合使用时的CI均大于1,即表现为拮抗作用。结论:牛蒡苷元与甲磺酸溴隐亭、牛蒡苷元与盐酸苯海索对鱼藤酮诱导的PC12细胞模型的无协同保护作用,反而呈现出一定的拮抗作用。Objective:To study the protective effects of arctigenin and verify the synergistic effect of combined arctigenin and positive drug(bromocriptine mesylate and benzhexol hydrochloride)on rotenone-treated PC12cells.Methods:Rotenone-treated PC12cells were employed for the in vitro experiments.The cells were exposed to rotenone with or without arctigenin and different positive drug solutions with different concentration.The optical density(OD)value was determined by microplate reade r,and the cell survival rate of each group was calculated.The cell viability was assessed using MTT to estimate the protective effect of arctigenin,positive drug only or combination of arctigenin and positive drug on the growth of PC12cells of different concentration,and the median-effect principle was used to analyze union of medication.Results:Compared to the model group,the cell viability of groups(arctigenin,bromocriptine mesylate and benzhexol hydrochloride)was significantly increased.According to the Chou-Talalay method,the combination index(CI)of arctigenin and bromocriptine mesylate(ratio:1∶4and1∶2)was beyond1,and the CI of arctigenin and benzhexol hydrochloride(ratio:1∶1and2∶1)was also greater than1,show that combination effect both were antagonistic action.Conclusion:Combination of arctigenin and bromocriptine mesylate or benzhexol hydrochloride have not synergistic protective effect on PC12cells treated with rotenone,but have antagonistic action.
