摘 要:目的观察芒果苷预处理对大鼠心肌缺血/再灌注损伤的作用。方法通过结扎冠脉30分钟再灌注2小时的方式制备心肌缺血/再灌注损伤模型,将大鼠随机分为5组:假手术组、模型组、芒果苷三个剂量组(40、20、10 mg/kg),于制备模型前7天开始灌胃给药;再灌注结束后,采用比色法检测血清肌酸激酶(creatine kinase,CK)活性、乳酸脱氢酶(lactate dehydrogenase,LDH)活性,染色法检测心肌梗死面积,定磷法检测心肌组织Na^+-K^+-ATP酶活力、Ca^(2+)-Mg^(2+)-ATP酶活力。结果与模型组(36.25%)比较,芒果苷高、中剂量组的心肌梗死面积分别缩小至32.44%和32.02%(P<0.05);与模型组(1137.26 U/L)比较,芒果苷各剂量组CK活性分别为907.31、899.20、848.65 U/L(P<0.05或P<0.01);与模型组(2345.82 U/L)比较,芒果苷中、高剂量组LDH活性分别为1722.54、1669.43 U/L(P<0.05);与模型组(4.22 mmol Pi/g Pro)比较,芒果苷高、中剂量组可显著升高Na^+-K^+-ATP酶活力为5.01、4.85 mmol Pi/gPro(P<0.05或P<0.01);高、中剂量组可显著升高Ca^(2+)-Mg^(2+)-ATP酶活力为6.12、6.19 mmol Pi/g Pro(P<0.05)。结论芒果苷预处理可保护心肌缺血/再灌注所致心肌损伤,机制可能与改善心肌组织的能量代谢有关。Objective To investigate the effects of mangiferin on energy metabolism in rats with myocardial ischemia/reperfusion injury.Methods SD rats were divided randomly into sham group,model group,mangiferin low dose group(10 mg/kg),mangiferin medium dose group(20 mg/kg)and mangiferin high dose group(40 mg/kg)(n=10).Mangiferin was orally administered once a day at 7 days before the modeling operation.Myocardial ischemia/reperfusion injury model was established by coronary artery ligation(30 min)and subsequent reperfusion(2 hours)in rats.After the modeling operation,the creatine kinase(CK)and lactate dehydrogenase(LDH)activities in serum were examined by chromatometry,Na+-K+-ATPase and Ca 2+-Mg 2+-ATPase activities in brain tissue were detected by phosphorus method and the myocardial infarction size(MIS)was determined by staining method.Results MIS of high and medium doses of mangiferin were significantly reduced to 32.44%and 32.02%,respectively,the difference was statistically significant compared with 36.25%of the model group(P<0.05).The CK activities in mangiferin groups were 907.31,899.20 and 848.65 U/L,respectively,which were significantly lower than 1137.26 U/L of model group(P<0.05 or P<0.01).LDH activities of high and medium doses of mangiferin were significantly reduced to 1722.54 and 1669.43 U/L,respectively,the difference was statistically significant compared to 2345.82 U/L of model group(P<0.05).Na+-K+-ATPase activity of Mangiferin(40 and 20 mg/kg)was increased to 5.01 and 4.85 mmol Pi/g Pro,respectively,the difference was statistically significant compared to 4.22 mmol Pi/g Pro of model group(P<0.05 or P<0.01).Ca 2+-Mg 2+-ATPase activity of Mangiferin(40 and 20 mg/kg)was significantly increased to 6.12 and 6.19 mmol Pi/g Pro,respectively,the difference was statistically significant compared to that of model group(P<0.05).Conclusion The protective effects of mangiferin pretreatment on myocardial ischemia/reperfusion injury may be related to the improvement of energy metabolism.
