摘 要:目的观察逍生散对干眼角膜上皮细胞损伤模型的干预作用,探讨逍生散治疗干眼的作用及分子作用机制。方法人角膜上皮细胞加入苯扎氯铵溶液制备得干眼角膜上皮细胞损伤模型,加入低、中、高剂量逍生散含药血清和空白对照血清孵育后,分别于作用24小时、48小时、72小时时行光镜观察,检测细胞活性、细胞凋亡及角膜上皮细胞中IL-1、IL-6、TNF-α、MMP-9细胞因子的表达情况。结果中药中、高剂量组能明显抑制细胞体积增大、细胞松解及间隙变大,减少细胞脱壁及溶解、坏死,减少细胞凋亡;中药中、高剂量的细胞活性值和其他组差异有统计学意义(P<0.05);中药中、高剂量组检测IL-1、IL-6、TNF-α及MMP-9的表达相对较低(P<0.05),且呈现随药物浓度增加,抑制炎症因子效果相对越好的结果。结论逍生散含药血清中、高剂量组能明显升高细胞活性;减少细胞凋亡;逍生散可能通过降低IL-1的产生,进而降低产生IL-6、TNF-α等炎症因子以及可能通过降低IL-1的产生,调控MMP-9的数量和活性,发挥保护角膜上皮细胞的作用。Objective To observe the intervention effect of Xiaosheng powder on corneal epithelial cell injury model,and to investigate the effect and molecular mechanism of Xiaosheng powder in the treatment of dry eye and.Methods Corneal epithelial cell injury model was established by human corneal epithelial cells joined Benzalkonium Chloride Solution.Low,medium and high dose of serum containing Xiaosheng powder and control serum containing blank was added after incubation.Light microscopy was used to observed at 24h,48h and 72h.Cell activity,apoptosis and the expression of IL-1,IL-6,TNF-αand MMP-9 cytokines in corneal epithelial cells were detected.Results The medium and high dose group can significantly inhibit cell volume enlargement,cell lysis and cell gap enlargement,reduce cell wall removal and dissolution,necrosis,and reduce cell apoptosis.There were significant differences between medium,high dose and other groups in the cell activity(P<0.05).The expression of IL-1,IL-6,TNF-αand MMP-9 in medium and high dose group was relatively low(P<0.05),and showed the better effect of inhibiting inflammatory factors with the increase of drug concentration.Conclusion Serum containing Xiaosheng powder medium and high dose group could significantly increase cell activity,induce apoptosis,decrease the production of IL-1,thereby reducing the production of IL-6,TNF-αand other inflammatory factors.It may play a role in protecting the corneal epithelial cells by reducing the production of IL-1 to regulate the number and activity of MMP-9.
