摘 要:通过临床症状观察、病理学检测、RT-PCR检测与序列分析等方法,对疑似患猪繁殖与呼吸综合征(Porcine reproductive and respiratory syndrome,PRRS)的病猪进行诊断。送检病猪临床表现发热、呼吸困难、皮肤发绀;剖检病变为肺脏实变、淋巴结肿大出血;组织学变化主要包括间质性肺炎、淋巴结出血及多核巨细胞浸润;免疫组化结果显示,猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)抗原主要定位于肺泡上皮细胞和巨噬细胞;病料RT-PCR结果呈PRRSV(命名为SJZ201701株)阳性;序列分析结果表明,该病毒的核衣壳(Nucleocapsid,N)蛋白基因与NADC30株之间基因核苷酸、氨基酸的同源性最高,分别为96.3%和84.4%。遗传进化分析表明,SJZ201701株与NADC30参考毒株关系最近。综合判定该病例为类NADC30株PRRSV引起的感染。The diagnosis of pigs suspected of having porcine reproductive and respiratory syndrome was carried out through clinical symptom observation,pathological examination,RT-PCR detection and sequence analysis.The clinical signs of the diseased pigs were fever,dyspnea and cyanosis of skin.At necropsy,consolidation of lung,swelling and hemorrhage of lymph nodes were observed.The pathological changes including interstitial pneumonia,hemorrhage and multinucleated giant cell infiltration of lymph node.The PRRSV antigen was mainly located in alveolar epithelial cells and macrophages by immunohistochemical staining.The RT-PCR result were positive for PRRSV,the virus was named SJZ201701 strain.Sequence analysis showed that the virus had the highest homology with the NADC30 strain.The nucleotide and amino acid sequence homology between the N protein gene of the two viruses were 96.3%and 84.4%respectively.Phylogenetic analysis showed the SJZ201701 strain had the closest relationship with NADC30 strain.The case was diagnosed as NADC30 like PRRSV infection.
