摘  要:目的:研究"三焦针法"对痴呆小鼠mPTP活性及Bcl-2蛋白的调控,以探究"三焦针法"对阿尔茨海默病神经元细胞凋亡的影响。方法:选取快速老化小鼠SAMP8为模型、以SAMR1为正常对照组,通过使用Western-blot、荧光分光光度计检测、TUNEL法等方法,观测各组小鼠皮质与海马的mPTP活性、Bcl-2蛋白含量变化、细胞凋亡情况及"三焦针法"对其影响。结果:SAMR1组小鼠皮质与海马mPTP活性较低,而SAMP8组小鼠皮质与海马mPTP活性增强,针刺组mPTP活性较SAMP8组相比明显降低。SAMP8组与SAMR1组比较,小鼠皮质与海马线粒体Bcl-2蛋白显著下调;针刺组与SAMP8组比较,bcl-2蛋白表达上调。SAMP8组小鼠皮质与海马的细胞凋亡情况与SAMR1组相比,有明显升高趋势;针刺组小鼠皮质与海马细胞凋亡情况较SAMP8组相比有下降趋势。结论:"三焦针法"具有调控SAMP8小鼠mPTP活性及抑制神经元细胞凋亡的作用。Objective:To study the effect of triple energizer acupuncture on the regulations of mPTP and Bcl-2 protein,thus to explore the influence of this therapy on neuronal apoptosis in Alzheimer disease(AD).Methods:The senescence-accelerated mice of SAMP8 were selected as the model,mice of SAMR1 were used as the normal control.mPTP activity,Bcl-2 protein content,and apoptosis of cortex and hippocampus were observed by using Western-blot method,fluorescence spectrophotometer detection,and TUNEL method respectively.The influence of the triple energizer acupuncture on them were assessed as well.Results:In terms of the activity of mPTP,it was lower in SAMR1 group than that in SAMP8 group,it became more significantly lower by acupuncture treatment compared to that in SAMP8 group.In terms of Bcl-2 protein expression,it was significantly down-regulated in SAMP8 group compared to that in SAMR1 group;however it was significantly up-regulated after acupuncture treatment compared to that in SAMP8 group.In terms of apoptosis,it had an increasing tendency in SAMP8 group compared to SAMR1 group;it had a decreasing tendency in acupuncture group compared to SAMP8 group.Conclusion:The therapy of triple energizer acupuncture can regulate activity of mPTP and inhibit cell apoptosis in SAMP8 mice.
