摘 要:目的探讨"芪茵颗粒"对非酒精性脂肪性肝病(NAFLD)模型大鼠肝组织中FATP4、L-FABP表达的影响及作用机制。方法雄性SD大鼠72只,随机分成空白对照组、模型组、芪茵颗粒低、中、高剂量组和阳性药物硫普罗宁组,每组12只。除空白对照组正常进食、自由饮水外,其余5组大鼠予高脂乳剂进行灌胃,每日1次,共10w,造模成功后分别用芪茵颗粒低、中、高剂量及硫普罗宁灌胃治疗,共10w。观察大鼠的一般情况,记录各组大鼠体质量,检测高密度脂蛋白(HDL-c)、低密度脂蛋白(LDL-c)、总胆固醇(TC)、甘油三酯(TG)和血清谷丙转氨酶(ALT)、谷草转氨酶(AST)等指标,采用Real-time PCR法检测肝组织FATP4、L-FABP mRNA的表达。结果与空白对照组相比,模型组大鼠LDL-c明显上升,HDL-c明显下降,TC、TG含量明显上升,血清ALT、AST含量显著升高,肝组织FATP4、L-FABP mRNA含量的表达增高,差异有统计学意义(P<0.05~0.01);经芪茵颗粒治疗后,与模型组相比,芪茵颗粒高、中剂量组LDL-c含量明显下降,HDL-c含量明显上升,TC、TG含量明显下降,血清ALT、AST水平明显降低,芪茵颗粒高、中、低剂量组肝组织FATP4、L-FABP mRNA含量的表达均降低,差异有统计学意义(P<0.05~0.01)。结论芪茵颗粒可以显著改善NAFLD大鼠脂质代谢紊乱,并能有效地降低大鼠血清LDL-c、TC、TG、ALT、AST水平及肝组织中FATP4、L-FABP mRNA的表达。ObjectiveTo investigate the effect and mechanism of Qi Yin granule on the expression of FATP4 L-FABP in rats with non-alcoholic fatty liver disease(NAFLD).MethodsAccording to the random distribution table,72 male SD rats were randomly divided into control group,model group,low,medium and high dose Qi Yin granule group and positive drug group(tiopronin),12 rats in each group.Except control group,the other 5 groups were given high fat emulsion by gavage,1 time daily for 10 weeks.When the model was established successfully,the rats were treated with low,medium and high dose Qi Yin granule,and tiopronin by gavage for 10 weeks,respectively.The general condition of rats was observed,the body weight and liver index were recorded,and the histopathological changes of liver were observed by light microscopy.The mRNA expression of FATP4 and L-FABP in liver tissue was determined by real-time PCR.ResultsCompared with the blank control group,the LDL-c of the model group was obviously increased and HDL-c was decreased,the content of TG was increased,and the content of ALT and AST was significantly increased in the model group.The mRNA expression of FATX4,L-FABP in liver tissue was significantly higher than that in control group(P<0.05).After treatment with Qiyin granule,compared with the model group,Qiyin granule was higher than that in the model group,the content of LDL-c in the middle dose group was obviously decreased and the content of HDL-c was obviously increased.The content of TCC-TG was obviously decreased and the serum ALT was obviously decreased in the middle dose group.The levels of AST and the mRNA expression of L-FABP in liver tissue of high,medium and low dose groups were significantly decreased.The difference was statistically significant(P<0.05).ConclusionQi Yin granule can control the growth of body weight in NAFLD rats,significantly improve the fatty degeneration of the liver index and hepatic tissue of NAFLD rats,effectively reduce the mRNA expression of hepatic FATP4 and L-FABP,and thus play a role in the tr
