摘  要:为研究番茄生长素运输载体SlPIN1对花器官脱落以及花器官中生长素分布的影响,利用病毒诱导基因沉默技术(VIGS)获得了pTRV-SlPIN1植株。20株pTRV-SlPIN1植株中,17株SlPIN1基因表达量下降,其中pTRV-SlPIN1-2和pTRV-SlPIN1-5植株表达量分别为对照pTRV植株的40%和35%。进一步研究pTRV-SlPIN1-2和pTRV-SlPIN1-5植株开花当日的番茄花器官发现,花柄离区至远轴端出现黄化,同时,田间花柄脱落率及离体花柄脱落率显著高于对照pTRV植株。通过对pTRV-SlPIN1及pTRV植株花器官进行GUS染色和生长素浓度检测发现,与对照相比,番茄SlPIN1基因沉默植株(pTRV-SlPIN1-2和pTRVSlPIN1-5)的花柄离区处生长素浓度显著下降,而子房基部、雄蕊和萼片等部位生长素浓度则明显上升,这不仅表明SlPIN1是番茄花器官中生长素运输的主要调控因子之一,其在生长素由花器官(源)向基部器官(库)方向运输过程中起重要作用,同时表明SlPIN1以此维持离区生长素的正常积累,来抑制番茄脱落的发生。本研究为番茄花器官中生长素极性运输的调控机理研究提供了参考,同时为SlPIN1及生长素极性运输调控脱落过程提供了依据。In order to investigate the role of SlPIN1,an auxin efflux carrier,in abscission and auxin distribution in tomato flower organ,we silenced SlPIN1 gene using a virus-induced gene silencing(VIGS)strategy.Among the 20 pTRV-SlPIN1 plants,the expression of SlPIN1 was suppressed in 17 plants compared to the control plants(pTRV),especially in pTRV-SlPIN1-2 and pTRV-SlPIN1-5(the transcripts of SlPIN1 gene were suppressed by 60%and 65%).We found that the flowers at anthesis of pTRV-SlPIN1-2 and pTRV-SlPIN1-5 silenced plants showed higher abscission rates than flowers of pTRV plants.Moreover,the flower pedicels gradually became yellow from abscission zone(AZ)to distal part.These might be due to the decreased auxin levels in AZ and increased auxin levels in ovary,stamen and sepal,respectively,which were observed in flowers of pTRV-SlPIN1-2 and pTRV-SlPIN1-5 by GUS staining and IAA quantification.These indicated that SlPIN1 has an important role in auxin source-sink transport in tomato flower organ,which is helpful for auxin maxima maintenance of AZ and prevention of abscission.The results provided a reference for mechanism research of auxin polar transport in tomato flower,and supported that SlPIN1 and auxin polar transport play roles in flower abscission process.
