摘  要:目的探讨炎症性声带疾病患者核转录因子kappa B(NF-κB)信号通路的表达变化。方法选取2015年6月-2017年1月在深圳市宝安区人民医院耳鼻咽喉头颈外科就诊的喉部慢性炎症患者(接受手术的声带息肉及小结患者)50例为疾病组,并选取50例正常声带组织(喉癌术中切取)标本作为对照组。HE染色观察手术患者声带病变组织是否发生炎症;ELISA检测TNF-α、IL-6、IL-1β的含量;实时荧光定量聚合酶链反应(q RT-PCR)和Western blot检测toll样受体4(TLR4)m RNA及蛋白的表达变化;Western blot检测NF-κB p65磷酸化水平。结果 HE染色示手术患者声带病变组织内发生炎症反应;ELISA结果显示:疾病组TNF-α、IL-6、IL-1β的含量,与对照组比较,差异有统计学意义(均P<0.05),疾病组均升高;疾病组TLR4 m RNA及蛋白的表达水平,与对照组比较,差异有统计学意义(P<0.05),疾病组均升高;疾病组NF-κB p65磷酸化水平,与对照组比较,差异有统计学意义(P<0.05),疾病组升高。结论 TLR4/NF-κB信号通路的活化在炎症性声带疾病中发挥作用。Objective To explore the expression changes of TLR4/NF-κB signal pathway in patients with inflammatory vocal diseases.Methods Fifty patients with chronic inflammation of the larynx who underwent vocal cord polyp and nodule surgery in our hospital from June 2015 to Janurary 2017 were chosen into the disease group and 50 cases of healthy vocal cord tissue(laryngeal carcinoma intraoperative removal)were selected as control group.HE staining was used to observe whether the vocal cord lesion of the surgery patients had inflammation.Enzymelinked immunosorbent assay(ELISA)was used to detect the content of TNF-α,IL-6 and IL-1βin the two groups.Quantitative real-time fluorescent PCR and Western blot were used to detect the expression changes of TLR4 mRNA and protein.Western blot was used to detect the phosphorylation level of NF-κB p65 in the two groups.Results HE staining showed inflammation in the vocal cord lesion of the surgery patients.ELISA results showed that the content of TNF-α,IL-6 and IL-1βof the disease group increased significantly,the differences were statistically significant compared with the control group(P<0.05).The expression levels of TLR4 mRNA and protein of the disease group increased significantly,the differences were statistically significant compared with the control group(P<0.05).The phosphorylation level of NF-κB p65 in the disease group increased significantly,the difference was statistically significant compared with the control group(P<0.05).Conclusions The activation of TLR4/NF-κB signal pathway plays a role in inflammatory vocal cord diseases.
