摘　　要：目的:观察smad4和cyclin A蛋白在体外培养大鼠卵巢颗粒细胞中的表达以及当归黄芪超滤膜提取物对其的增殖作用。方法:以不同剂量的当归黄芪超滤膜提取物作用于体外培养的大鼠卵巢颗粒细胞,用MTS法检测各组大鼠卵巢颗粒细胞24、48和72小时的增殖情况,并用westren bolt法监测smad4和cyclin A蛋白的表达。结果:smad4和cyclin A蛋白在颗粒细胞高表达,随着细胞增殖作用的增加,其表达也逐渐增加;当归黄芪超滤膜提取物可促进颗粒细胞的增殖,并且呈一定的量效和时效关系,最佳作用浓度为0.375g/L;各组比较差异有统计学意义(P<0.05)。结论:当归黄芪超滤膜提取物有促进卵巢颗粒细胞增殖的作用,能促进卵泡发育,与上调smad4和cyclin A蛋白表达有关。Objective:To observe the expressions of smad4 and cyclinA protein in ovarian granulosa cells cultured in vitro of the rats and the mitogenic effects of the extract of DangGui(Angelica sinensis)and HuangQi[Astragalus membranaceus(Fisch.)Bunge.]ultrafiltration membrane on their expressions.Methods:After acting on ovarian granulosa cells cultured in vitro of the rats with different concentrations of the extracts of DangGui and HuangQi ultrafiltration membrane,the proliferation of the granulosa cells in different groups in 24 hours,48 hours and 72 hours was detected by MTS method,the expressions of smad4 and cyclinA protein were monitored by western blot method.Results:The expressions of smad4 and cyclinA protein were in high levels in granulosa cells,as the proliferation of cells increased,its expressions raised gradually,the extracts of DangGui and HuangQi ultrafiltration membrane could promote the proliferation of granulosa cells,which demonstrated certain dose-effect and time-effect relationship,the optimal acting concentration was 0.375g/L,and the difference had statistical meaning(P<0.05).Conclusion:The extracts of DangGui and HuangQi ultrafiltration membrane could stimulate the proliferation of ovarian granulosa cells and promote the development of the follicle,which might be related to upregulating the expressions of smad4 and cyclinA protein.
