摘  要:以大叶千斤拔总黄酮含量为指标,考察大孔树脂、聚酰胺树脂、硅胶柱层析及溶剂萃取法对大叶千斤拔中总黄酮的富集效果。结果表明,乙酸乙酯萃取法和硅胶柱层析法联用能有效富集大叶千斤拔中的总黄酮。最佳纯化工艺为:大叶千斤拔提取液浓缩至无醇味,用2倍体积的乙酸乙酯萃取5次,所得萃取液浓缩蒸干后,用甲醇溶解至0.1 g/m L,拌样,上200-300目硅胶柱,上样量为1∶50,径高比为1∶15,用氯仿-甲醇(18∶1,v/v)的洗脱液洗脱,根据薄层结果,合并黄酮部分,浓缩蒸干后即得大叶千斤拔的黄酮提取物,纯度为54%。With the total flavonoids content ofMoghania macrophylla as indexes,the enrichment effect of macroporous resin,polyamide resin,silica gel column chromatography and solvent extraction method were compared.The combination usage of ethyl acetate extraction and silica gel column chromatography could enrich the total flavonoids of Moghania macrophylla effectively.The optimal purification process are as follow:The extracting solution of Moghania macrophylla was concentrated to no alcohol flavor,extracted five times with 2 times the volume of ethyl acetate;the extracted solution was dehydrated,dissolved in methanol to 0.1g/mL,the sample was mixed and loading to silica gel column of 200-300 mesh,the sampling amount was 1:50 and the diameter to height ratio of the column was 1:15,the eluent component was chloroform and methanol with the ration 18:1,the flavonoids was merged according to the results of the thin-layer chromatography and then dehydrated,the flavonoids concentration of Moghania macrophylla was 54%.
