摘  要:目的研究藏药灰兜巴的不同提取物对Ⅱ型糖尿病肾病(DKD)模型小鼠肾损伤的保护作用。方法实验通过STZ配合饲喂高糖高脂饲料诱导DKD小鼠模型,给予灰兜巴水提物、醇提物以及多糖不同提取物高(200 mg/kg)、低(100 mg/kg)剂量8周后,对小鼠血肌酐(SCr)、血尿素氮(BUN)、尿蛋白(mg/24 h)、糖化血清蛋白(Gsp)、终末糖基化产物(AGEs)等指标以及肾脏病理组织学改变进行检测分析,研究藏药灰兜巴对DKD的改善作用,并对肾脏损伤相关蛋白纤维连接蛋白(FN)、细胞间粘附因子(ICAM-1)、转化生长因子(TGF-β1)进行Western blot检测与分析。结果藏药灰兜巴不同提取物可降低小鼠血清血糖及AGEs含有量,降低相关靶点蛋白的表达,有效缓解肾小球基底膜增厚,系膜基质增多,肾小管、上皮细胞变性等病理状态,改善DKD小鼠肾功能损伤。结论藏药灰兜巴3种提取物均可改善Ⅱ型DKD小鼠肾脏损伤状态。AIM To study the efficacy of different extractions of Huidouba,a Tibetan medicine to mice with type IIdiabetic kidney disease(DKD).METHODS The type II DKD mice models induced by the injection of streptozotocin(STZ)on base of high glucose and high fat diet subsequently treated with different extractions of Huidouba were studied to investigate the protective effects of Huidouba on renal injury.The mice were given high dose(200 mg/kg)and low dose(100 mg/kg)of Huidouba ethanol extract,Huidouba water extract and Huidouba polysaccharide respectively for 8 weeks.Their serum creatinine(SCr),blood urea nitrogen(BUN),urine protein(mg/24 h),glycosylated serum protein(Gsp)and advanced glycation end products(AGEs),their histological changes of renal pathology were detected and analyzed.Meanwhile their protein levels of fibronectin(FN),intercellular adhesion factor(ICAM-1)and transforming growth factor(T G F-pl)were identified by Western blot.RESULTS The different extractions of Huidouba,demonstrating a portfolio of capacity in decreasing levels of serum glucose and AGEs,inhibiting the expression of target protein,alleviating the glomerular basement membrane thickening,improving mesangial matrix,renal tubular and epithelial cells degeneration and other pathological conditions,improved the renal function of DKD mice.CONCLUSION The three extractions of Huidouba highlight the renal improvements in type II DKD mice.
