摘 要:目的探讨泽兰甲醇提取物对慢性软组织损伤模型大鼠的影响。方法采用机械冲击挫伤造模,正常喂养2周复制慢性软组织损伤大鼠模型,然后连续每天1次灌胃泽兰甲醇提取物高、中、低剂量14 d,在给药的第1、2、3周末,通过测定受损肌肉组织中超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、前列腺素E2(prostaglandin E2,PGE2)、一氧化氮(nitric oxide,NO)及血清中白介素-6(interleukin-6,IL-6)及组胺水平、肿瘤坏死因子(tumor necrosis factor-alpha,TNF-α)、Ⅰ/Ⅲ胶原蛋白(Collagen-I/Ⅲ)表达以及血管内皮生长因子(vascular endothelial growth factor,VEGF)活性、内源性碱性成纤维细胞生长因子(basic fibroblast growth factor,b FGF)活性,探讨泽兰甲醇提取物对慢性软组织损伤大鼠模型的影响及其机制。结果泽兰甲醇提取物可明显降低肌肉组织中MDA、PGE2及NO含有量和TNF-α、IL-6和组胺水平(P<0.05),显著升高肌肉组织中SOD(P<0.01),可显著抑制组织中Collagen-I/Ⅲ蛋白的表达(P<0.01),早期显著提高组织中VEGF和bFGF活性(P<0.01)。结论泽兰甲醇提取物对慢性软组织损伤模型大鼠具有一定的治疗作用,其机制与抑制损伤部位的炎症因子水平,改善其氧化应激,并与抑制Ⅰ/Ⅲ胶原蛋白过度表达,增强VEGF和bFGF活性加快修复速度有关。AIM To investigate the effect and mechanism of methanolic extract of Eupatorium(MEOE)to model rats with chronic soft tissue injury.METHODS The model rats were established by mechanical injury and a subsequent two-week normal feeding for respective administration of high,medium and small dosage of MEOE once a day successively for 14 days.An array of indices,the level of superoxide dismutase(SOD),malondialdehyde(MDA),prostaglandin E2(PGE2),nitric oxide(NO),interleukin-6(IL-6)and histamine,the expression of tumor necrosis factor-alpha(TNF-a),nitric oxide(NO)and Collagen-I/IH,and the activity of vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF)were measured to analyze the effect of MEOE to model rats with chronic muscle injury.RESULTS MEOE resulted in apparent reduction of contents of MDA,PGE2 and NO,and the levels of TNF-a and IL-6 in muscular tissue(P<0.05),significantly increased of the SOD in muscular tissue(P<0.01),a remarkably inhibited expression of the tissue Collagen-I/IH protein(P<0.01),and significantly improved activity of tissue VEGF and bFGF(P<0.01).CONCLUSION The certain therapeutic effects of MEOE to rats with chronic muscle injury may correlate with its influence to the levels of inflammatory factors inhibition,the oxidative stress relief,the overexpression of collagen-I/IH inhibition,the VEGF and bFGF activity improvement,and the time spare from the repairing.
