摘　　要：为分析宠物犬源大肠杆菌对常用抗菌药物的敏感性,从洛阳市宠物医院采集宠物犬肛门拭子样品,获得了3株犬大肠杆菌。通过麦康凯琼脂培养基和伊红美蓝琼脂选择性培养基,对菌株进行了分离培养。采用煮沸法提取大肠杆菌DNA,采用PCR方法扩增大肠杆菌16S r DNA。用琼脂糖凝胶电泳检测了PCR产物大小,并对扩增产物进行了测序,用BLAST软件进行同源性对比分析,最终鉴定为大肠杆菌。选择20种抗菌药物,采用纸片扩散法进行药敏试验。试验结果表明:宠物犬源大肠杆菌对氨苄西林、庆大霉素、阿莫西林、环丙氟哌酸等16种抗菌药物出现不同程度的耐药性,且耐药谱广,出现了多重耐药。In order to analyze the sensitivity of Escherichia coli( E. coli) isolated from pet dogs to commonly used antimicrobial agents,anal swab samples from pet dogs in Luoyang pet hospital were collected to obtain three dog E. coli. The strain was isolated and cultured by Mac Conkey agar medium and eosin methylene blue agar selective medium. The E. coli DNA was extracted by boiling method,and 16S rDNA of E. coli was amplified by polymerase chain reaction( PCR) method. The size of PCR products were detected by agarose gel electrophoresis and the amplification products were sequenced. The homology of E. coli was compared and analyzed by BLAST software,and the strain was identified as E. coli. The Kirby-Bauer method was used to carry out drug sensitivity test of selected 20 antimicrobial agents. The results show that E. coli isolated from pet dog can resist to 16 kinds of antibacterial drugs,such as ampicillin,gentamicin amoxicillin and ciprofloxacin. Wide drug resistance spectrum and multiple resistant bacteria are present.
