黄芪虫藤饮对类风湿关节炎大鼠关节滑膜细胞生长增殖的影响

摘　　要：目的探讨黄芪虫藤饮对类风湿关节炎大鼠关节滑膜细胞增殖的影响。方法无菌条件下分离正常大鼠和类风湿关节炎(RA)模型大鼠关节滑膜组织,制成单个滑膜细胞,流式细胞术鉴定后,取培养第三代的关节滑膜细胞进行药物干预,用MTT法测定药物对细胞增殖抑制作用;用流式细胞术检测细胞凋亡率以及细胞周期。结果正常组大鼠关节滑膜细胞增长率与RA空白组(0.00 mg/L)比较,差异有统计学意义(P<0.05);不同浓度(100.0,50.0,25.0,12.5,6.25,0 mg/L)黄芪虫藤饮对RA大鼠关节滑膜细胞增殖均有明显的抑制作用,细胞凋亡率明显高于空白组(P<0.05),并能诱导滑膜细胞G0/G1期的细胞生长停滞,且呈现典型的剂量依赖性。结论黄芪虫藤饮能抑制RA关节滑膜细胞的活性,促进细胞凋亡,可以较好地应用于临床。Objective To study the effect of Huangqi Chongteng drink on the proliferation of synovial cells in rats with rheumatoid arthritis.Methods The synovial cells of normal rats and RA model rats were isolated under aseptic condition to prepare single synovial cells.Flow cytometry was used to identify the third synovial synoviocytes.The inhibitory effect of the drug on cell proliferation was measured by MTT assay.The apoptotic rate and cell cycle were measured by flow cytometry.Results The growth rate of synovial cells in the normal group was significantly higher than that in the blank group(0.00 mg/mL)(P<0.05).Different concentrations of Huangqi Chongteng drink(100.0,50.0,25.0,12.5,6.25,0 mg/mL)had obviuos inhibition effect on the proliferation of synovial cells in RA rats,and the cell death rate was significantly higher than that of the blank group(P<0.05).The cell growth rate of G0/G1 phase in synovial cells was significantly higher than that in the blank group.Conclusion Huangqi Chongteng drink can inhibit the activity of synovial cells in RA model rats and promote cell apoptosis,which can be applied in clinical practice.
